Objective:idiopathic short stature(ISS)means that the cause of the disease is unknown and that recombinant human growth hormone(rhGH)is the first choice for treatment.Although the dose of rhGH in children with ISS is relatively large,the clinical effect of children is different,and some children with Idiopathic dwarfism have poor response to treatment.In order to further clarify the etiology and guide the treatment of ISS and under the normal levels of the serum growth hormone(GH),insulin-like growth factor binding protein-3(IGFBP-3)and Insulin-like growth factor-1(IGF-1)in childrens,we investigate the levels of serum IGFALS protein and IGFALSmRNA in childrens between ISS and normal.We will find the pathogenesis of idiopathic short stature,which provided a theoretical basis for finding the pathogenesis and guiding treatment of ISS.Methods:A total of 26 children with idiopathic short stature were admitted to this study as ISS group,and 10 age-matched normal height children were selected as control group.The comprehensive medical history and physical examination results were collected.Serum IGFALSmRNA expression and IGFALS protein level were measured by RT-PCR and ELISA way.T-test,correlation analysis and other statistical methods were used to analyze the results.Results:1.The serum IGFALS protein expression level and IGFALSmRNA expression showed no significant difference between ISS group and control group(P>0.05).2.The levels of IGFALS protein was negatively correlated with height difference(r=-0.729,P=0.000)(P<0.05),and IGFALSmRNA was negatively correlated with height difference(r=-0.475,P=0.000)(P<0.05);The levels of IGFALS protein and IGFALSmRNA were no both significant difference correlated with age?sex and BMI(P>0.05).3.IGFALS protein expression was positively related to IGFALSmRNA expression in ISS children(P<0.05).Conclusions:1.The decrease of IGFALS protein is related to the height of ISS children,and it can be one of the etiology of children with idiopathic short stature.2.In this study,under the condition of normal growth hormone,IGFALSmRNA transcription was not affected in ISS group and control group,while IGFALSmRNA was negatively correlated with height difference in ISS group,which had clinical significance.3.IGFALS mRNA in ISS group is positively related to IGFALS protein,which is consistent with the transcription and translation of the gene level,and the conclusion further supports that the reduction of IGFALS protein may be the cause of some patients with idiopathic short stature.
