Objective: Pancreatic cancer(PC)is one of the most common malignant tumors of the digestive system.Curcumin is the main component extracted from the rhizome of the ginger family.In recent years,many studies have proved that curcumin can play an anti-tumor and pro-cancer role in a variety of tumor cells,including human pancreatic cancer cells.It is known from literature that YAP/TAZ gene is involved in the regulation of organ development and is highly expressed in various human tumor cells such as liver cancer and breast cancer.Therefore,in this study,the effects of different concentrations of curcumin on the growth of human pancreatic cancer cell lines(Patu8988 and MIA paca-2)were studied,and whether the expression of YAP/TAZ in these two cell lines was affected,and the possible mechanism of curcumin in human pancreatic cancer was preliminarily discussed.Methods:1.CCK-8 method and plate cloning experiment were used to detect the effects of different concentrations of curcumin on the proliferation inhibition and cloning formation ability of PC cells;2.PI staining with flow cytometry was used to detect the effects of curcumin at different concentrations on the apoptosis of PC cells,and the changes of cell morphology were observed under the microscope.3.The migration ability of the two kinds of cells was detected by scratch test with different concentrations of curcumin.4.Western blot was used to detect the effect of curcumin treatment on the expression of YAP and TAZ protein in the two kinds of cells 48 h after treatment.Results:1.CCK-8 method showed that curcumin significantly inhibited the proliferation of PC cells,and the effect was concentration-dependent.The results of plate cloning experiment showed that compared with the control group,the ability of cell cloning formation was decreased in curcumin treatment group,and proliferation was significantly inhibited.2.The PI staining with flow cytometry showed that curcumin could induce apoptosis,and its effect was enhanced with the increase of drug concentration.Under the microscope,it was observed that curcumin could cause morphological changes in cells and the extent of the changes was proportional to the drug concentration.3.Scratch test results showed that different concentrations of curcumin inhibited the migration ability of PC cells.The inhibitory effect was enhanced with the increase of drug concentration.4.Western blot results showed that curcumin significantly inhibited the expression of YAP/TAZ proteins in PC cells.Conclusion:Curcumin can inhibit the proliferation of Patu8988 and MIA paca-2,reduce the ability of clone formation,promote apoptosis,inhibit cell migration.The mechanism may be related to down-regulating the expression of YAP and TAZ genes in cells.
