Objective:In recent years,thymoma and EBV-related tumors are the hotspots and focuses of research,but fewer studies can link these two,and related mechanisms are still unclear.This study was designed to investigate the differences in EBV infection and immunoregulatory expression levels in patients with type B thymoma,and to explore the relationship between these immunoregulators and EBV infection in thymoma.At the same time,comparison of different types,clinical stages and MG with EBV infection and immunoregulatory factors in thymoma samples.It'll provide a basis for further study of the possible mechanism of EBV in thymoma and will provide a new way for the prevention and treatment of thymoma.Methods:In this experiment,immunohistochemical staining was used to detect the expression of LMP1 in B-type thymoma.We studied the relationship between LMP1 expression in B-type thymoma and EBV infection and the relationship between thymoma and myasthenia gravis by differently grouping the results.At the same time,we collectedserum samples from the same patient,through ELISA experiment,to test EBV infection in the sample,meanwhile,we contrasted with the results of immunohistochemistry to confirmtheir relationship.In addition,we selected focus tissue samples from patients who was with subtypes of type B thymoma,extracted total RNA,reverse transcribed cDNA furtherly and tested the expression of EBV-related immunomodulatory factorsin B-type thymoma tissues by Real-time PCR,including CCL25,SH2D1 A and MYC genes,to explore the differences in the expression levels of these immunomodulatoryfactors in thymoma tissue.Results:The results of immunohistochemistry showed that LMP1 was widely expressed in thymoma tissues,and its expression was significantly correlated with the occurrence and clinical stages of thymoma.The positive rate of LMP1 was stage III>I stage>II stage,which had statistical significance(P< 0.05),and had no obviously statistically significant difference with MG(P>0.05);the ELISA method for EBV infection in peripheral blood showed that the EBV positive rate in the simple thymoma group was higher than that with the MG thymoma group,which had statistical significance(P<0.05),and the level of EBV infection in peripheral blood was not significantly different from the occurrence of thymoma and stages of thymoma(P>0.05);Real-time PCR showed that CCL25 was more up-regulated in thymoma than normal thymus,which had statistical significance(P<0.05),and the expression level of CCL25 was related to the pathological type of thymoma,the expression level of B1 thymoma was higher than other types,which had statistical significance(P<0.05),and there is no significant correlation between the expression level of CCL25 and whether patients have MG.(P>0.05).There was no significant difference in the expression levels of SH2D1 A and MYC between B-type thymoma and normal thymus tissue,there was also no significant correlation of expression levels between different clinical stages of thymoma and between simple thymoma and with myasthenia gravis(P> 0.05).Conclusions:The expression of LMP1 in thymoma tissues was significantly correlated with the occurrence and clinical stage of thymoma,EBV infection was associated with the occurrence of thymoma.EBV may aggravate the invasiveness and even the malignancy of thymoma.The expression of LMP1 was not significantly associated with the presence or absence of MG.Peripheral blood EBV infection was associated with MG in patients with thymoma,but there was no significant correlation with pathological type and stages of thymoma.The up-regulated expression of CCL25 is related to the occurrence of thymoma and pathological type of thymoma,it may that the activation of this immunomodulatory factorsparticipate in the pathogenesis of thymoma,but the expression of CCL25 is not related to MG;there was no significant correlation betweenthe expression of SH2D1 A,MYC and the occurrence of thymoma,stages of thymoma,pathological type and MG.
