PurposeThe subject studies Liraglutide Intervention in different doses on rats with cerebral ischemia reperfusion.The experiment is to observe the changes on inflammatory factors of brain tissue at different times after reperfusions.The essay analyzes the mechanism of reactions on inflammation after its restrains against cerebral ischemia reperfusion.The research provides data and theoretical foundations for Liraglutide clinical application and relative animals experiments.Methods72 healthy male rats,weigh 180-220 grams,were selected as samples.They were grouped in random,into surgery group,model group,Liraglutide Intervention group with high dose,and Liraglutide Intervention with low dose.MCAO draws a model of reperfusion,2 hours after middle cerebral artery occlusion.Neurology scoring system is applied for evaluating rats at the point of 6 hours,12 hours,24 hours and 7 days respectively after reperfusion.TTC Staining determines the capacity of rats cerebral infarction,HE Staining detects neuronal degeneration and necrosis.Immunohistochemical method examines the content of TNF-? and IL-6.In sit hybridization tests the conveys.ResultsEvaluation of neurologic impairment: Sham-operation control group did not find impairment on neurologic scoring,while it appears in other three control groups.Among them,model control group has relatively severe neurologic impairment.Compared with model control group,Liraglutide in both high and low doses control groups have remarkable lower degree of neurologic impairment at the time points of 6 hours,12 hours,24 hours and 7 days(P<0.05).The significance differences also exist among Liraglutide groups with different doses.Evaluation of TTC staining determining the capacity of rats cerebral infarction: There is no cerebral infarction in Sham-operation control group,while other four groups have higher possibility of cerebral infarction(P<0.05).Compared with model group,the volume of cerebral infarction in Liraglutide intervention groups decreased at 12 hours and 7days(P<0.05).Volumes of cerebral infarction in Liraglutide intervention groups are quite different.HE Staining: 6 hours after cerebral ischemia reperfusion,karyopyknosis of nerve cell can be discovered under optical microscope.The shape changes,but there is no significant edema around neuron.24 hours,7days after reperfusion,the effect of impairment becomes significant,large amount of stained neuron nucleus in dark can be discovered.Nucleus are intensed and solid.Most structures of cells disappear,and edemas are clear.The evaluation of TNF-? immunohistochemical tests.Compared with Sham-operation control group,other groups show remarkably positive on TNF-?(P<0.05).Compared with model control group,Liraglutide intervention control groups show clear decrease at the point of 12 hours and 24 hours7days(p<0.05),significant differences also appear in different Liraglutide intervention control groups with different doses.The evaluation of IL-6 immunohistochemical tests: Compared with Sham-operation control group,all control groups have significant difference on positive IL-6 expression(P<0.05).Compared with model control group,Liraglutide intervention control groups show lower positive expressions at the 24 hours,7days(p<0.05);Significant differences also appear in different Liraglutide intervention control groups with different doses.In situ hybridization TNF-? mRNA: Compared with model control group,positive expression of Liraglutide intervention control groups with small dose drops significantly at 12 hours 24 hours and 7days(P<0.05).Compared with model control group,positive expression of Liraglutide intervention control groups with large dose drops significantly at 6 hours,12 hours and 7 days(p<0.05).The rate of decreases for each Liraglutide intervention control group are quite different.The expression of IL-6 mRNA immunohistochemical tests: Compared with model control group,expression of IL-6 mRNA in Liraglutide intervention control groups with small dose drops significantly at 12 hours,24 hours and 7days(P<0.05).Compared with model control group,expression of IL-6 mRNA in 7Liraglutide intervention control groups with large dose drops significantly at 6 hours,12 hours,24 hours and 7days(p<0.05).The rate of decreases for each Liraglutide intervention control group are quite different.ConclusionThe analysis confirms the influence of Liraglutide on mechanism of inflammatory response to cerebral ischemia-reperfusion injury on rats.After the application of Liraglutide Intervention,the expression rates decrease significantly at different testing point based on the brain tissues TNF-??IL-6 of rats in cerebral ischemia.In addition,neurological function is expected to be improved,the volume of cerebral infarction is expected to decrease.Intervention control group gets more significant results.
