Research purpose:1.A total of 265 prescriptions for essential hypertension treated by Professor Liu Zhongyong were collected.The association rules and potential drug combinations were analyzed by data mining method of association rule and complex system entropy clustering.2.According to the combination of data mining and network pharmacology,the mechanism of the treatment of essential hypertension was deeply understood.Research method:Based on the software of traditional Chinese Medicine inheritance Assistance system(V2.5),this paper studies the research method of entropy clustering of complex systems by collecting Professor Liu Zhongyong's prescriptions for the treatment of essential hypertension,using association rules.Then the network pharmacological mechanism of drug combination was excavated by Cytoscape3.6.1 software(http://www.cytoscape.org/).1.Data mining analysisProfessor Liu Zhongyong's prescriptions for the treatment of essential hypertension were collected and screened,analyzed as data sources,and found out the core combinations and new prescriptions,so as to clarify the internal relationship between "disease-prescription and medicine".At the same time,analysis of symptoms,signs and drug usebetween the law.2.Network pharmacological analysisTaking the association rule Apriori algorithm and the entropy clustering method of complex system as the research object,using the method of network pharmacology to construct the compound-target network and target interaction PPI network,in order to explore the salvia miltiorrhiza and Ligusticum chuanxiong.Pharmacological mechanism of Poria-Atractylodes in the treatment of essential hypertension.The interaction between chemical components and target proteins in four Chinese herbs and the signal pathway for the treatment of this disease were discussed.Research results:1.Study on prescriptions of Professor Liu Zhongyong for treating essential Hypertension based on data MiningIn the total prescription study,the drugs used in the first six times of frequency were Ligusticum chuanxiong(207),Salvia miltiorrhiza(199),Radix Atractylodes(146),Poria cocos(139),Rhizoma ternata(135),Atractylodes macrocephala(133);The top five drug combinations of high frequency were "Ligusticum chuanxiong-Danshen(158)","Salvia miltiorrhiza(144)","Ligusticum chuanxiong(119)","ChuanxiongDanshen-Muxiang(118)","Atractylodes macrocephala-Poria cocos(115)";When the number of support is more than 53 and confidence is more than0.9,there are 77 association rules of drug combination,of which 23 have confidence of 1.Twelve drug core combinations were obtained based on entropy clustering of complex systems.Based on unsupervised entropy hierarchical clustering,six new prescriptions were obtained.2.Research on prescription symptoms and signs of Professor Liu Zhongyong in the treatment of essential Hypertension based on data MiningIn the overall prescription study,the symptoms and signs of the first five times of frequency were dizziness(135),chest tightness(98),tongue redness(92),dry mouth(82),fatigue(81).The first five symptoms and signs were "dizziness-chest tightness(59)","dizziness-dry mouth(58)","dizziness-tongue pale red(52)","asthenia-dizziness(51)","chest tightness-palpitation(48)";When the number of support was more than 12 and confidence > 0.7,there were 43 association rules of symptom and sign combination,among which the first five were "more dream-> dizziness(0.9375)"."Arthralgia-> dizziness(0.93333)","palpitations,tongue dark red-> chest tightness(0.875)","dry mouth,silky veins-> dizziness(0.8666)","swelling,palpitations-> chest tightness(0.8571)".3.Study on the Action Mechanism of Salvia miltiorrhiza RhizomaThe chemical constituents of Radix Salviae Miltiorrhizae and Ligusticum chuanxiong were searched by BATMAN-TCM database,and the target of drug components was screened out,including 166 compounds of Salvia miltiorrhiza and 465 potential targets,while Ligusticum chuanxiong contained 182 compounds and 955 potential targets.The targets collected from Salvia miltiorrhiza and Ligusticum chuanxiong were mapped to essential hypertension targets in DisGeNET database,including 234 targets of Salvia miltiorrhiza and 252 targets of Ligusticum chuanxiong.The chemical constituents of Poria cocos and Atractylodes macrocephala were searched and the targets of drug components were screened out.Among them,Poria cocos contained 54 compounds with 503 potential targets,and Atractylodes macrocephala contained 26 compounds and 56 potential targets.The target of Poria cocos and Atractylodes macrocephala collected was mapped to the target related to essential hypertension,including 141 of Poria cocos and 18 of Atractylodes macrocephala.Research conclusion:1.Based on the data mining of traditional Chinese Medicine(TCM)inheritance Assistant system,this study analyzed Professor Liu Zhongyong's medical records of treating essential hypertension(vertigo),and summarized Professor Liu Zhongyong's academic thought and clinical experience in the treatment of essential hypertension.2.Radix Salviae Miltiorrhizae-Bulbus Fritillariae Cirrhosae and Poria-Rhizoma Atractylodis Macrocephalae have multiple chemical components,multiple targets and multiple channels.The Danshen-Chuanqi drug is likely to act on the high blood pressure via signal pathways such as cAMP,calcium ion,cGMP-PKG,HIF-1,TNF and prolactin.It is possible to play a role in hypertension by the pathway of cAMP,cytokine-factor receptor interaction,calcium ion,cGMP-PKG,TGF-1 and so on.
