Objective: To observe the effects of heat-sensitive moxibustion“Zhongwan”(CV12)acupoint on serum growth hormone(GH),pepsinogen(PG)and gastric mucosa c-myc,survivin mRNA in Rats with CAG,to explore the possible mechanism of heat-sensitive moxibustion for treating CAG,so it can provide part of experimental basis for clinical treatment.Methods: To meet the requirements of 66 animals were randomized:blank group and model group.The rats in the blank group did not intervene,the rats in the model group were treated with the composite etiology model method to establish CAG model for 12 weeks,they were randomly divided into the model group,vitacoenzyme group and moxibustion group,the rats in the moxibustion group were bound and moxibustion at “Zhongwan”for 40 min,after moxibustion,2ml/kg/d was treated with intragastric administration of 0.9% Nacl,according to the change of tail temperature during moxibustion,the rats were divided into heat-sensitive moxibustion group and non heat-sensitive moxibustion group.The rats in the vitacoenzyme group were treated with identical volume of vitacoenzyme after binding 40 min.The rats in the model group were treated with identical volume of 0.9% NacL after binding 40 min.Both once a day for28 days.The blank group received no stimulation during treatment.The rat general condition,body mass were observed in each group.After the treatment,6 rats were randomly selected from each group,the expressionsof serum GH,PG were detected by enzyme linked immunesorbent assay(ELISA),the expressions of gastric mucosa c-myc,survivin mRNA were detected by Real time Quantitative polymerase chain reaction(QPCR)method,the histopathological changes of gastric mucosa were observed by optical microscope.Result:(1)Before modeling,all the observation indexes(baseline),such as general condition and body mass of rats in each group were similar.After model establishment,the rats in the blank group were in good condition,the body mass increased normally.The rats in the model group were dispirited,activity slower,diet decreased,hair was withered,back was weaked,the skin mucosa was pale and matte,with dim ear and Pale tail,the stool was soft or watery.The body mass of model group was significantly lower than the blank group,and the difference was statistically significant between them(P<0.01).After modeling,there was no obvious abnormality in the gastric mucosa of the blank group observed by naked eye and optical microscope.The gastric mucosa of the rats in the model group showed dark red or gray color by naked eyes,the mucosa layer became thinner,elasticity decreased,plica became flat,mucus decreased,tendency was disorder,gully became shallow or disappeared,and the muscular layer became thinner.Under the light microscope of the gastric mucosa of the rats in the model group,the gastric mucosa layer was obviously thinner and hyperemic,the lamina propria glands was atrophy or even disappeared,some of the gastric glands was cystic dilation,lymphoid follicles and inflammatory cells or heteromorphic hyperplasia was observed.(2)Compared with the blank group before treatment,the body mass of rats in other groups were significantly reduced(both P<0.01).Compared with the model group before treatment,there was no significant differencebetween the non heat-sensitive moxibustion group,heat-sensitive moxibustion group and vitacoenzyme group.After treatment,the body mass of rats in each group were significantly increased compared with that before treatment(both P<0.01).After treatment,the body mass in the model group still significantly different compared with the blank group(P<0.01).Compared with the model group after treatment,the body mass in the non heat-sensitive moxibustion group,heat-sensitive moxibustion group and vitacoenzyme group were increasing obviously(both P <0.05).However,there was no significant difference among the three.After treatment,the general condition of rats in each group was improved.Under light microscope,there was no obvious abnormality in the gastric mucosa of the blank group,while in the model group,the gastric mucosa became thinner,the glandular cavity expanded while the glands atrophied,and intestinal metaplasia was occasionally observed.The other groups were improved compared with the model group,especially the heat-sensitive moxibustion group.(3)After the end of treatment,the levels of serum GH,PG?,PG? and PGR ratio in the model group were significantly reduced than the blank group(P<0.05,P<0.01).Compared with the model group,the levels of serum GH,PG ?,PG ? in the non heat-sensitive moxibustion group,heat-sensitive moxibustion group and vitacoenzyme group were remarkably increased(P<0.05,P<0.01),PGR ratio was no significantly increased.The heat-sensitive moxibustion group in increasing serum GH,PG?levels were superior to the non heat-sensitive moxibustion group and vitacoenzyme group(both P<0.05).(4)After the end of treatment,compared with the blank group,the expressions of gastric mucosa c-myc and survivin mRNA in the model group were significantly increased(both P < 0.01).The non heat-sensitive moxibustion group,heat-sensitive moxibustion group and vitacoenzymegroup could all reduce the expressions of c-myc and survivin mRNA in gastric mucosa compared with the model group(both P<0.01).Both the heat-sensitive moxibustion group and vitacoenzyme group were superior to the non heat-sensitive moxibustion group(P < 0.05,P < 0.01).The vitacoenzyme group was superior to the heat-sensitive moxibustion group in reducing c-myc mRNA(P<0.05).The heat-sensitive moxibustion group and vitacoenzyme group have similar effect in reducing survivin mRNA.Conclusion:(1)Heat-sensitive moxibustion can significantly improve the general conditions of CAG model rats(such as mental state,stool characteristics,etc.),body mass and the pathological morphology of gastric mucosal tissue.(2)Heat-sensitive moxibustion can increasing the serum levels of GH and PG?,to changes the atrophic state of gastric mucosa in rats with CAG.(3)Heat-sensitive moxibustion can inhibit the expressions of c-myc,survivin mRNA in gastric mucosa of rat model,restore the balance between proliferation and apoptosis of gastric mucosa cells and achieve the purpose of treating CAG.
