Objective To discuss the effect of Qingzao Jiufei Decoction on Key rate-limiting enzymes and intermediate metabolites of pentosephosphate pathway,Krebs cycle in lung cancer,and then explore the influence and action mechanism of Qingzao Jiufei Decoction on lung cancer.Method 50 male C57BL6J mice were randomly divided into five group:model group,CTX group,and high dose group,medium dose group,low dose group of Qingzao Jiufei Decoction.Animal models were induced with the axillary injection of Lewis cells,the Qingzao Jiufei Decoction group started medication at molding two weeks before(11,5.5,2.75g·kg-1·d-1),CTX group(50mg·kg-1,QOD)and model group?same volume of saline?started medication after molding.After 14d modeling,killed the mice and weigh tumor.We detected the enzymatic activity of Glucose-6-Phosphate Dehydrogenase?G6PD?,content of reactive oxygen species?ROS?,Cholesterol and Free Fatty Acids?FFA?by Elisa method.We detected the protein expression level of Aconitase 1?ACO-1?protein by Western-blot method.We detected the expressions of gp91phox and p22phox mRNA,which are subunit unit of NADPH oxidase and ATP citrate?pro-3S?-lyase?ACL?mRNA by Realtime-PCR method.Result1.Effect of the Qingzao Jiufei Decoction on tumor weight of Lewis-bearing miceCompared with the model group,the tumor weight of CTX group,high dose group,medium dose group and low dose group of Qingzao Jiufei Decoction were decreased obviously,the difference was significant?P<0.01?.Compared with the CTX group,the tumor weight of high dose group,medium dose group and low dose group of Qingzao Jiufei Decoction were increased obviously,the difference was significant?P<0.01?.Compared with the high dose group of Qingzao Jiufei Decoction,the tumor weight of medium dose group and low dose group of Qingzao Jiufei Decoction were increased obviously,the difference was significant?P<0.01?.Compared with the medium dose group of Qingzao Jiufei Decoction,the tumor weight of low dose group of Qingzao Jiufei Decoction were increased obviously,the difference was significant?P<0.05?.The tumor inhibition rate of CTX group,high dose group,medium dose group and low dose group of Qingzao lung cancer cells.and inhibit the energy metabolism.3.Qingzao Jiufei Decoction maybe can inhibit the expression of ACO-1 and ACL,reduce the supply of tricarboxylic acid cycles to cellular synthetic materials FFA and CHO,and inhibit the energy metabolism.
