Objective Sulinglong-1 jianxiu is a herbs extracts preparation prepared by Yishou Peiyuan(Guangzhou)Health Management Co.Ltd.for obese people.It is mainly composed of extracts from Cassia seed,mulberry leaf and other herbs.Its pharmacological effects are not clear.This study established a rat obesity model to study its effect on obesity and explore its possible mechanism.Method Obesity rat model was established by feeding high fat diet for 10 weeks.Orlistat(0.044g/kg/d)was used as positive control.The rats were treated with Sulinglong-1 jianxiu of 0.738,1.476,2.952g/kg/d by gavage administration respectively for 42 days.During the course of treatment,the rats were fed with corresponding feed.They were weighed once a week.42 days later,the rats were anesthetized with ether.Their body lengths were measured.Their blood samples were collected from femoral artery.Their liver tissue,fat tissue and cecum contents were collected for the determination of relevant indicators.The contents of total cholesterol(TC),triglyceride(TG),low density lipoprotein cholesterol(LDL-C),high density lipoprotein cholesterol(HDL-C),free fatty acid(FFA),lipid lipase(LPL)and hepatic lipase(HL)in the serum were determined with corresponding kits,and the contents of adiponectin(ADPN)in the serum of rats were measured with ELISA.Obesity index(Lee's index)and fat-body ratio were calculated.The pathological changes of liver tissue and fat tissue were observed with hematoxylin-eosin(HE)staining.The DNA samples of cecum content were extracted.16 S rRNA gene PCR was carried out to analyze the composition of intestinal flora.In addition,rat jejunum homogenate was prepared to detect the inhibitory effect of the sample on intestinal mucosal ?-glycosidase.The inhibitory effect of the sample on the absorption of sugar in rats,pancreatic beta cells of which were damaged with alloxan,was further tested.Result After 10 weeks of modeling,The animals with a body weight of 20% higher than the average body weight of normal group were used as model animals.42 animals were successfully modeled and the success rate was 52.5%.After the treatment,compared with the model control group,Sulinglong-1 jianxiu can significantly reduced the body weight,obesity index,perirenal fat,genital fat,total fat and fat body ratio(P<0.01 or P<0.05).And can significantly reduce serum TC,TG,LDL-C,FFA content(P<0.01 or P<0.05),and can significantly increase serum HDL-C and ADPN levels and LPL,HL activity(P<0.01 or P<0.05).Pathological observation showed that the liver fatty lesions of the rats in each administration group were significantly relieved compared with the model control group,and no obvious pathological changes were observed in the hepatocytes,the diameter of adipocytes in each administration group was smaller,and the number of adipocytes in the same field of vision under the microscope was significantly increased.These results indicate that Sulinglong-1 jianxiu has a good effect on obesity and can improve liver damage caused by high fat under no change in feeding.The results of researching mechanism showed that Sulinglong-1 jianxiu could inhibit sugar absorption in rats by inhibiting ?-glucosidase activity(P<0.05).And it can improve disorder of intestinal flora caused by high fat diet in rats.At the phylum level,compared with the model control group,the number of Firmicutes decreased and Bacteroidetes increased significantly(P<0.05)and returned to normal level.At genus level,the number of Lactobacillus,Bacteroidales_S24-7_norank increased significantly.The abundances of Desulfovibrio and Ruminococcaceae_unclassified were significantly reduced(P<0.01 or P<0.05).Conclusion Sulinglong-1 jianxiu can reduce the weight of obese rats,improve lipid metabolism,relieve liver fatty lesions under no change in feeding.It has a good effect on obesity.Its mechanism of action may be related to inhibition of polysaccharide absorption by inhibiting ?-glucosidase.It may also play a role in the treatment of obesity to increase the abundance of probiotics,to reduce the abundance of harmful bacteria in the intestine,and adjusting the micro-ecological balance of the intestine.This can provide a theoretical basis for the study of the new pharmacological effects of Sulinglong-1 jianxiu on obesity.
