The Expression Of Rac1 And Its Regulation Of Biological Function In Gastric Cancer

Objection:1)To investigate the expression of Rac1 in gastric cancer and its relationship with clinicalpathological characteristic and prognosis of gastric cancer patients.2)To investigate the role of Rac1 in gastric cancer tumorigenesis and metastasis.Methodes:1)The expression of Rac1 in 92 human gastric cancer were detected by immunohistochemistry;2)The correlation of Rac1 expression with clinicopathological characteristics in gastric cancer was analyzed by Pearson's?~2analyses;3)The correlation of Rac1 expression with survival of gastric cancer patients was analyzed by Kaplan-Meier analyses;4)The biological behaviors of SGC-7901/Rac1 and SGC-7901/Vector were evaluated by CCK-8 assay,soft agar assay and transwell assay;5)In vivo,the correlation of Rac1 expression and transplantation tumor growth in nude mice was analyzed.Results:1)Rac1 is highly expressed in gastric cancer;2)Positive expression of Rac1correlated with differentiation,local invasion,lymph node metastasis,and Lauren's classification(all P<0.05);3)Rac1 overexpression in gastric cancer predicted a shorter survival;4)In vitro study showed that in gastric cancer cells,overexpression of Rac1 could promote cell proliferation,migration and invasion(all P<0.05),and inhibition of Rac1 by specific inhibitor could abrogate these effects(all P<0.05);5)In vivo,overexpression of Rac1 could promote the growth of transplantation tumor in nude mice.Conclusion:These findings demonstrated that Rac1 was highly expressed in gastric cacner and was correlated with the clinicopathological characteristics and was a factor for poor prognosis.In addition,experimental models supported the role of Rac1 in promotion of gastric cancer cell growth,migration and invasion.Given its pivotal role in tumorigenesis and progression,Rac1 could be served as a promising therapeutic targets for gastric cancer.
