Objective:Examining the difference in expression of Retinol Binding Protein 4 in placenta between the gestational diabetes mellitus pregnant women and normal pregnant women to explore its relation to the pathogeneses of GDM.Methods:Placental tissues were collected from pregnant women who received regularly care in Shanghai General Hospital from June 2015 to January 2016..27normal pregnant women were selected as the control group.26 pregnant women diagnosed as GDMA1,which have well blood glucose without insulin 26 pregnant women diagnosed as GDMA2,which have well blood glucose by insulin.The expression of RB P4 in placenta was determined by immunohistochemistry,Western Blot and quantitative real-time PCR.Results:1.The result of IHC.RBP4 is located on the cytoplasm of trophoblastic cells.The final scores of IHC in the normal,GDMA1and GDMA2 are 3.04±1.58 vs4.37±2.09vs4.43±2.04.The differences among the three groups are statistically significant?P<0.05?.The differences between GDMA1and GDMA2 have no statistically significant?p>0.05?.2.The result of WB.The ratio of RBP4/GAPDH in the normal,GDMA1and GDMA2are 1.27±0.67vs2.38±0.69 vs2.82±0.91.The differences among the three groups are statistically significant?P<0.01?.The differences between GDMA1and GDMA2 have no statistically significant?p>0.05?3.The result of qRT-PCR.The 2-??CT in the normal,GDMA1and GDMA2 are 1.84±2.44 vs8.16±2.90 vs8.76±6.92,.The differences among the three groups are statistically significant?P<0.05?.The differences between GDMA1and GDMA2 have no statistically significant?p>0.05?Conclusions:The expression of RBP4 in placenta is increased in GDM,however,there is no statistically significant between GDMA1and GDMA2.The increased expression of RBP4 in placenta may associate with the pathogenesis of GDM,however,there is no proof display the relation to the serious degree of GDM.
