| It has been studied that intestinal flora is an important bridge between the host and the environment and plays a role in the host’s health.Probiotics have been shown as an effective adjuvant therapy to improve insulin resistance.The aim of this paper is to explore the promotion of probiotics on hypoglycemic function of acarbose in diabetic mice and the possible mechanism,and to propose a new idea for diabetes therapy.Probiotics’utility of carbon sources,intestinal colonization ability,the inhibition activities on intestinal opportunistic pathogens and interaction with acarbose were investigated in this paper.Lactobacillus acidophilus HCCB20535,Lactobacillus rhamnosus HCCB20555,Bifidobacterium longum HCCB20558 and Bacillus licheniformis HCCB20544 among 9 strains were selected as composition of hypoglycemic agent with acarbose finally.The effect of tetragenus probiotics on diabetes prevention in mice was studied.The mice were fed with diet enriched with high glucose and high fat,then 4×109cfu of tetragenus probiotics were administrated intragastrically every day for 3 weeks.It was found that the weight of mice fed with probiotics was significantly lower than that of mice fed with HFD only.And compared to the control group,daily consumption of tetragenus probiotics reduced the rate of streptozotocin(STZ)–induced diabetic mice in probiotic group to 42.9%.So tetragenus probiotics had preventive effect on diabetes.After low-dose tetragenus probiotics(4×109cfu/day)was administered to mice for 21 days,the promotion of tetragenus probiotics to acarbose on hypoglycemic function was tested.Neither tetragenus probiotics nor 30 mg/kg of acarbose alone showed obvious hypoglycemic activity in mice.However,postprandial blood glucose reduced significantly when 30 mg/kg of acarbose combined with tetragenus probiotics was administrated.Plasma triglyceride(TG)of tetragenus probiotics group was significantly lower than that of diabetic group,while the probiotics group had a much higher GLP-1 level.Besides,taking probiotics could modulate the gut microbial dysbiosis of diabetic mice,an increase in the abundance of Lactobacillus and a decrease of bacteroides and Clostridium.As well as the level of acetic acid was reduced in probiotics group.Intragastrical administration of high-dose tetragenus probiotics(2×1010cfu/day、1.2×1011cfu/day)for one time could also improve the hypoglycemic function of acarbose.Tetragenus probiotics could promote acarbose to reduce postprandial blood glucose in diabetic mice.It is possible to combine the tetragenus probiotics with acarbose as an effective adjuvant in diabetes therapies. |
