Objective: in order to further clear after process optimization of Mongolian wood of shiny-leaved yellowhorn adminstration of analgesic anti-inflammatory effect,the first adopts ultraviolet spectrophotometer,choose the reasonable by the method of orthogonal experiments of wood of shiny-leaved yellowhorn alcohol extraction process of total flavonoids,polymer materials together again in the splendid alkene acid sodium(glue),gansu aminoacetate(cross-linking agent),tartaric acid(PH regulator)and glycerin(moisturizer)as the substrate preparation of gel preparation.The matrix formula was optimized by the star-point design-response surface method(CCD),and the analgesic and anti-inflammatory effects of Mongolian wood of shiny-leaved yellowhorn plaster were observed by hot plate instrument,acetic acid torsion method and xylene ear swelling method.Methods: experiment 1.Extraction process of total flavonoids from arborvitae bunge: in this study,ethanol reflux extraction method was used to extract the effective components of arborvitae bunge,and the optimal extraction process was determined by orthogonal experiment.Among them,the single factor experiment was carried out for ethanol extraction.Investigation factors and levels: according to the relevant literature reports,the investigation factors of alcohol extraction process of guanyu wenguanyu are: ethanol concentration,ethanol dosage,extraction time and extraction times.Three levels are designed for each factor.First,the optimal concentration of ethanol and two boundary values are determined by single factor test.Then make the orthogonal test factor level table.The experiment was arranged according to the experimental conditions designed in the orthogonal design table L9(34).Finally,the best extraction technology was determined.Experiment 2,optimization of matrix prescription of wen guan mu plaster by the star-spot design effect surface method: through the preliminary test,the influence of the amount of drug,the order of drug and matrix addition,adhesive,moisturizer,cross-linking agent regulator and other factors on the molding of wen guan mu plaster was investigated to screen out the suitable prescription of plaster.The star Design-response surface method(CCD),and select initial adhesion force,adhesion force and film residue as the evaluation index,NP-700,nucleoside aminoacetate,glycerin,tartaric acid dosage to examine factors,using four factors,five levels of star Design to arrange the prescription,preparation of 31 groups of gel preparation,center repeated test seven times,the Design-Expert model fitting the data,according to the test results of optimizing prescription matrix.Experiment 3.Extraction process of myricetin from arbutus arbutus: orthogonal test was conducted to determine the content of myricetin by HPLC,and the best extraction process of myricetin was selected.Combined with the extraction process of total flavonoids and myricetin,the final extraction process was obtained.Experiment 4: skin irritation and irritability test of wen guan mu plaster: 1.Skin redness and blisters were used as observation items to evaluate the irritability of wen guan mu plaster.2.Optimized wenguanmu plaster for skin allergy test.Erythema and blisters were used to observe asthma,standing instability and shock in mice.Scoring method was used to determine whether wenguanmu plaster had anaphylaxis in mice.Experimental 5: study on the anti-inflammatory and analgesic effects of wenguanmu plaster: the analgesic effects of wenguanmu plaster were observed by writhing body method in mice and hot plate method in mice.To observe the anti-inflammatory effect of wen guan mu plaster by ear swelling method in mice.Conclusions: this study discussed the extraction technology of total flavonoids wood of shiny-leaved yellowhorn rhizome,on this basis,explore the wood of shiny-leaved yellowhorn of arbutus pigment content in roots,the results showed that the ethanol refluxing extraction of total flavonoids extracted wood of shiny-leaved yellowhorn optimum process is: the reflux extraction time is 2 h,70% ethanol concentration,solid-liquid ratio 1:25(g/m L),extraction times for 2 times,this condition below crown wood rhizome yield of total flavonoids was 8.5467%.According to the variance results,ethanol concentration,extraction time and extraction times are the three main factors affecting extraction efficiency.Secondly,HPLC method was used to determine the content of myricetin in rhizome.The optimal extraction process of myricetin was as follows: the ethanol concentration was 60%,the solid-liquid ratio was 1:30,and the extraction time was 1h.According to the comprehensive extraction process,the optimal extraction process of total flavonoids can contain 93.81% of the total content of myricetin.Therefore,the conclusion is that the extraction process of total flavonoids can be used for the extraction of effective components from the rhizome of Chinese arborvitae.Moreover,the ethanol reflux extraction method has the advantages of simple process,convenient operation,low cost and no environmental pollution,etc.,which will provide a theoretical reference for the development and utilization of rhizome.Progresses in the extraction process of foundation style wooden traditional paste formulations reform research,through thestar design response surface method,the conclusion is: when the 4.00 g,polyacrylic acid sodium aminoacetate 0.60 g,tartaric acid is 0.20 g,glycerin and ml,peppermint oil,0.5 ml,turpentine 0.5 ml,borneol 0.02 g is the highest score,at the time of the paste forming good,meet the requirements of viscosity.Irritability and irritability were found in normal skin during skin irritation test and allergy test.The irritant of damaged skin is 0,no irritant.In the study on analgesic and anti-inflammatory effects of wenguanmu plaster,the analgesic effect of wenguanmu plaster observed by hot plate instrument was as follows: compared with the blank group,the positive control group and wenguanmu medium dose group had significant analgesic effects(P<0.05).Compared with the traditional group,the medium dose group had significant analgesic effect(P<0.05).In the study on the analgesic effect of the mice torsion method,the incubation period test results showed that: compared with the blank group,the positive control group had significant analgesic effect(P<0.05).;Compared with the positive control group,the traditional group and the plaster group had significant analgesic effect(P<0.05).There was no significant difference between wenguanmu traditional paste group and wenguanmu gel paste group.Compared with the positive control group,there were significant analgesic effects in wenguanmu traditional group and wenguanmu gel paste group(P<0.05).Compared with the traditional group,there was no significant difference between the two groups.6.In the study of xylene-induced ear swelling in mice to observe the anti-inflammatory effect of wenguanmu plaster,compared with the blank group,the traditional wenguanmu plaster group and the high-dose wenguanmu plaster group had significant anti-inflammatory effect(P<0.05),while no significant anti-inflammatory effect was found in the positive control group.Compared with the traditional group,the low-dose,medium-dose and high-dose wenguanmu plaster group had significant anti-inflammatory effects(P<0.05).The reformed wenguanmu ointment was used for skin irritation and allergy test,as well as analgesic and anti-inflammatory test.The test results were ideal,and the problems such as poor viscosity,dirty clothes and poor transdermal property were optimized on the basis of maintaining the effect of traditional wenguanmu ointment,so as to make it more close to the needs of patients in life.
