| Objective Mongolian medicine Oxytropis myriophylla herb is the dry aerial part of the Oxytropis myriophylla(Pall)DC.It is widely distributed in Heilongjiang,Liaoning,Inner Mongolia,Hebei,Shanxi,Ningxia and other places.Mongolian medicine Oxytropis myriophylla herb is commonly used drugs in the clinic,through market research,we found that many Mongolian doctors have self-use for the self-contained of Oxytropis myriophylla herb,and its quality standards is uneven.So far,there has been no report on the quality of Oxytropis myriophylla herb.In this study,a systematic and comprehensive phytochemical study of Oxytropis myriophylla herb was carried out,and the content of flavonoids was determined.Under the premise of identifying the main secondary metabolites of Oxytropis myriophylla herb,the fingerprints were studied at the same time.To provide a scientific basis for clarifying the pharmacodynamic basis of Oxytropis myriophylla herb and establishing quality evaluation methods.Methods 1.The ethanol extracts of Oxytropis myriophylla herb were separated and purified by atmospheric pressure silica gel column chromatography,HP-20 adsorption resin,MCI adsorption resin,Sephadex LH-20 column chromatography,etc.,and identification of the structure of compounds using modern spectral techniques;2.Fingerprint of 10 batches of Oxytropis myriophylla herb from different producing areas HPLC,and the common peaks were calibrated;3.Rutin was used as a reference substance for the first time,and the content of total flavonoids in Oxytropis myriophylla herb was determined by UV spectrophotometry,and the extraction conditions were optimized.Results 1.A total of 19 compounds were obtained by phytochemical separation,and the structure of 13 was identified.Other compounds were being identified.Among the identified compounds,7 flavonoids were included: kaempferol-7-O-α-L-rhamnopyranoside(1),quercetin-7-O-α-L-rhamnopyranoside(2),quercetin-3-O-(6-trans-caffeoyl)-β-glucopyranosyl-(1→2)-β-glucopyranoside-7-O-α-rhamnopyranoside(3),kaempferol(4),quercetin(5),linarin(7),diosmin(9);2 sterols: β-sitosterol(11),daucosterol(8);3 Phenolic acids: salicylic acid(10),protocatechuic acid(12),gallic acid(13);and D-3-O-methyl inositol(6).2.Fingerprints of 10 batches of Oxytropis myriophylla herb from different producing areas were established by HPLC,and 21 common peaks were calibrated.3.For the first time,the content of total flavonoids in 16 batches of Oxytropis myriophylla herb was determined by using rutin as reference substance,and the extraction conditions were optimized.The results showed that the total flavonoids content of 16 batches of Oxytropis myriophylla herb was 8.596 ~ 36.01mg·g-1.Conclusions 1.Compounds 1~3,6,9,10,12 and 13 were isolated from Oxytropis myriophylla herb for the first time.2.HPLC was used to establish the fingerprint of Oxytropis myriophylla herb,which is simple in operation and reproducible.It can comprehensively and objectively reflect the integrity of the multi-leaved Oxytropis myriophylla herb.3.The UV spectrophotometry operation is simple and quick,the equipment requirements is easy,good stability and reproducibility,and it can be used for the determination of the total flavonoids of Oxytropis myriophylla herb.This study systematically studied the quality of Oxytropis myriophylla herb,provided a scientific basis for the quality evaluation of Oxytropis myriophylla herb,and provided scientific theoretical and material basis for subsequent pharmacological activities and other related experiments. |
