Objective The polymorphisms of SLCO1B1 and AopE loci were observed in Mongolian patients with coronary heart disease and healthy people in Ordos region.The distribution of SLCO1B1 and AopE loci polymorphisms in Mongolian population in Ordos region was analyzed at the molecular level.SLCO1B1388A>G was analyzed.And the relationship between 521T>C,ApoE 526C>T and 388T>C SNP sites and coronary heart disease,provide a theoretical basis for the prevention and treatment of coronary heart disease and individualized treatment of clinicians.Methods Using the case-control method,four polymorphic loci of SLCO1B1*1b,SLCO1B1*5,ApoE2 and ApoE4 were detected by LightCycler480 real-time PCR.Using SPSS22.0 statistical software,the genotype frequencies of SNP loci in coronary heart disease group and control group were calculated by frequency calculation method.Results Four ApoE genotypes were detected in 100 patients with coronary heart disease diagnosed by CAG or CTA: ApoE2/E3,ApoE3/E3,Apo E3/E4,and ApoE4/E4.Among them,9 cases(9%)of ApoE2/E3 genotype,70 cases(70%)of ApoE3/E3 genotype,19 cases(19%)of ApoE3/E4 genotype,and 2 cases of ApoE4/E4 genotype(2 cases)%);the distribution frequency of alleles was ApoE3(84%),ApoE4(11.5%)and ApoE2(4.5%).There was no significant difference in ApoE genotype distribution between CHD group and control group(P>0.05).The frequency of ?2 allele in CHD group was lower than that in control group(P <0.05),and the frequency of ?3 allele was higher than that in control group(P <0.05).There was no significant difference in the frequency of the ?4 allele.The TGA concentration of the ApoE3/3 genotype group in the CHD group was higher than that in the control group,while the HDL-C,Lpa,and ApoA-1 levels were significantly lower than those in the control group(P<0.05).The levels of HDL-C and ApoA-1 in the ApoE3/4 genotype group of CHD group were lower than those of the control group,and the concentration of APOB was higher than that of the control group(P<0.05).There was no difference in the distribution of ApoE gene polymorphism between different sexes.Seven phenotypes of the SLCO1B1 gene were detected in the CHD group*15/*15,*1a/*15,1a/*1a,*1a/*1b,*1a/*5,*1b/*15 and *1b/ *1b,the proportion is 1%,5%,4%,26%,0%,13%,51%.The normal metabolites of SLCO1B1 inthe CHD group accounted for 81%,intermediate metabolites and weak metabolites,respectively,at 18% and 1%.The biochemical parameters of different phenotypes of CHCO group and control group SLCO1B1 gene were compared: the TGA level of 1a/15 genome in CHD group was higher than that of control group and HDL-C level was lower than that of control group,the difference was statistically significant(P<0.05);CHD The levels of ApoA-1 and HDL-C in group 1a/1b were lower than those in the control group(P<0.05).The levels of CHOL,TGA,LDL-C and APOB in group 1b/15 of CHD group were higher than those in control group.The difference was statistically significant.The TGA level of the1b/1b genome in the CHD group was higher than that in the control group,and the ApoA-1and HDL-C levels were lower than the control group,P<0.05.There was no significant difference in the frequency distribution of SLCO1B1 gene phenotype between CHD group and control group.There was no difference in the frequency distribution of SLCO1B1 genotype between different genders.Logistic regression model analysis showed that age,diabetes,smoking,ApoA-1,TGA were positively correlated with coronary heart disease.Conclusion The ApoE3 alleles were most widely distributed in the CHD group and the control group,with the highest number of patients with ApoE3/E3 coronary heart disease and the second with ApoE3/E4.The 2 allele of ApoE is a protective factor for coronary heart disease,and the 3 allele is a susceptibility gene for coronary heart disease,and the 4 allele is not associated with coronary heart disease.ApoE gene polymorphism is closely related to blood lipid levels.The frequency of distribution of APOE and SLCO1B1 gene polymorphisms was not related to gender.In the coronary heart disease group,SLCO1B1 mutations accounted for 19%,ApoE3/E4 and ApoE4/E4 genotypes accounted for 21%.These patients had poor dose tolerance to statins,prone to adverse drug reactions,and poor lipid-lowering effects.The risk of cardiovascular and cerebrovascular diseases is significantly increased.Therefore,it is particularly important to screen APOE and SLCO1B1 gene polymorphism before using statins to treat cardiovascular and cerebrovascular diseases.
