Purpose: Establish the microbial limit test method for Mongolian medicine Ji Rug-6 suppository to provide data for the establishment of its quality standard.To study the antibacterial and fungal effects of Mongolian medicine Ji Rug-6 suppository in vitro.Method:According to the fourth edition of the 2015 edition of the Chinese Pharmacopoeia,the applicability experiment of microbial limit test was carried out on the Mongolian medicine Ji Rug-6 suppository.The total number of aerobic bacteria,the total number of fungi and yeast counts,and the control bacteria were all diluted.Clinical and Laboratory Standards Institute(CLSI)CLSI-M100(K-B test)was used to test the effect of Mongolian medicine Ji Rug-6 suppository on staphylococcus aureus and escherichia coli,and CLSI-M27-A3(K-B test)was used to test the bacteriostatic effect of Mongolian medicine Ji Rug-6 suppository on candida albicans.The minimal inhibitory concentration of Mongolian medicine Ji Rug-6 suppository against staphylococcus aureus and candida albicans was determined by double dilution method.Result:Microbial limit test: When the dilution concentration of Mongolian medicine Ji Rug-6 suppository is 1:1000,the recovery rate of aerobic bacteria,mold and yeast is 0.63-1.23;the recovery rate of mold and yeast is 0.95-1.13.Staphylococcus aureus,Pseudomonas aeruginosa,Candida albicans and Clostridium sporogenes were detected in the bacterial liquid verification group of the control bacteria test.No growth was observed in the test group and the negative control group.qualified.In vitro antibacterial experiment: When the concentration of Mongolian medicine Ji Rug-6 was 1g/ml,the inhibition zone of Staphylococcus aureus was 8.7mm,and the inhibition zone of Candida albicans was 9.5mm,which showed no antibacterial effect on escherichia coli.When the concentration of Mongolian medicine Ji Rug-6 powder was 1g/ml,it showed no antibacterial effect on staphylococcus aureus,escherichia coli and candida albicans.When the concentration of Ji Rug-6 powder was 2g/ml,the bacteriostatic zone of staphylococcus aureus was 9.0mm,and there was no bacteriostatic effect on escherichia coli and candida albicans.The minimum inhibitory concentration of Mongolian medicine Ji Rug-6 supposition on staphylococcus aureus was 250mg/ml,and the minimum inhibitory concentration of candida albicans was 125mg/ml.Summary:According to the dilution method of pharmacopoeia of the People's Republic of China(2015 edition),microbial limit test was conducted on Mongolian medicine Ji Rug-6 suppository.The Mongolian medicine Ji Rug-6 suppository is stronger than Ji Rug-6 powder in its bacteriostatic effect on staphylococcus aureus.For the bacteriostatic effect on candida albicans,Mongolian medicine Ji Rug-6 suppository showed effect,while Ji Rug-6 powder showed no effect.Mongolian medicine Ji Rug-6 suppository and Ji Rug-6 powder showed no bacteriostatic effect on escherichia coli.
