Objective?1?In order to clarify the basis of the pharmacodynamics of Ferula ferulaeoides,this study carried out systematic phytochemical analysis and Ferula ferulaeoides clarified its main chemical substances;?2?Establish a fingerprint and quantitative analysis method for the medicinal materials of Ferula ferulaeoides.Methods?1?Separated and purified of monomeric compounds in Ferula ferulaeoides by atmospheric pressure silicagel column chromatography,Sephadex LH-20 column chromatography,reverse phase ODS column chromatography,TLC and liquid chromatography,the compounds were identified by UV,ESI-MS,1H-NMR,13C-NMR and other spectroscopy techniques,physical and chemical methods;?2.1?HPLC was used to study the fingerprints of the Ferula ferulaeoides.By comparing with the retention time of the reference materials,some chemical components in the fingerprints were identified;?2.2?Determinated ferulic acid,latifolone,guaiol and lehmannmlon in Ferula ferulaeoides with HPLC;?2.3?Determinated total coumarin of Ferula ferulaeoides by 7-hydroxy coumarin as indicator component with UV.Results?1?29 compounds were isolated from 70%ethanol extract of Ferula ferulaeoides,and 11 compounds were identified,namely:umbelliferone?1?,lehmannmlone?2?,guaiaol?3?,latifolone?4?,2,4-dihydroxyacetophenone?5?,dshamirone?6?,2,4 dihydroxybenzoic Acid?7?,methyl 2,4-dihydroxybenzoate?8?,ethyl 2,4-dihydroxybenzoate?9?,?-sitosterol?10?,daucossterol?11?.Among them,compounds 1,2 and 6 were isolated from ferula ferulaeoi-des for the first time,and compounds 8 and 9 were isolated from plants of the genus Ferula.?2.1?HPLC fingerprints of 10 batches of Ferula ferulaeoides were established from different producing areas,28 common peaks were identified.8 common chemical components were identified that:ferulic acid,2,4-dihydroxyacetophenone,Latifolone,lehmannmlone and dshamirone et.al;Similarity degree of the fingerprints concerning 10 batches of Ferula ferulaeoides were>0.9;?2.2?Concentration of ferulic acid,latifolone,guaiol and Lehmannmlone with the peak showed a good linear relationship respectively in the range of 0.12401.2400?g,0.09050.9050?g,1.355013.5500?g,0.0713?g0.7128?g,??=0.9996,?=0.9998,?=0.9998,?=0.9993?,and the average recoveries rate were 102.61%,96.39%,100.19%and 102.11%respectively,RSD is less than 2.05%;?2.3?7-hydroxycoumarin showed a good linear with the absorbance value in the range of 4.114.3?g,??=0.9999?,the average recovery rate was 101.82%,and the RSD was1.59%.Conclusions Through the study of the chemical constituents of Ferula ferulaeoides,its chemical base was further clarified.The fingerprint of Ferula ferulaeoides established by HPLC was representative and could reflect the medicinal materials integrity comprehensively;the quantitative analysis method of the multi-umbrella ferulide component established by HPLC is accurate,reliable and reproducible.The content of the total coumarin of Ferula ferulaeoides is simple,accurate and reliable determined by UV,and its quality of Ferula ferulaeoides has significant reference value.
