Expression Of MicroRNAlet-7a In Serum In Laryngeal Cancer And Its Significance

Objective: To investigate the changes of serum miRNAlet-7a expression in serum in laryngeal squamous cell carcinoma(LSCC)patients and the diagnostic value of miRNAlet-7a for laryngeal squamous cell carcinoma.Method: qRT-PCR was used to detect the expression of serum miRNA let-7a in 42 cases of laryngeal carcinoma,adjacent tissues,serum samples(before and after surgery)and 42 healthy subjects.The correlation of serum levels of miRNAlet-7a and laryngeal squamous cell carcinoma was analyzed.The relationship was analyzed by the receiver operating curve(ROC)for its diagnostic efficacy against laryngeal squamous cell carcinoma.Result:(1)The relative expression of miRNAlet-7a in the normal tissues adjacent to the cancer was(1.066±0.191)vs(0.241±0.138),and there was a significant difference between the groups(t=22.690,P<0.01).The relative expression of serum miRNAlet-7a in experimental group and the normal control group was(0.38±0.0831)vs(0.933±0.0723),and the difference was significant(t=32.536,P<0.01);The relative expression of serum miRNAlet-7a in preoperative and postoperative laryngeal cancer patients within 1 week was(0.38±0.0831)vs(0.488±0.1468),and the difference was significant between the groups(t=4.149,P<0.01).(2)The relative expression of miRNAlet-7a in cancer tissues was significantly different between different TNM stages and lymph node metastasis(t=1.819,0.234;P<0.05).The relative expression of serum miRNAlet-7a was associated with lymph node metastasis(P<0.05);it was not associated with age,gender,alcohol consumption,smoking,depth of invasion,tumor type,TNM stage,and degree of differentiation(P>0.05).(3)The relative expression level of miRNAlet-7a in laryngeal cancer tissues was correlated with that in serum(r=0.412,P=0.000).(4)The degree of 0.765 was regarded as the optimal threshold of the relative expression of serum miRNAlet-7a for the diagnosis of laryngeal cancer,of which the sensitivity was 76.5%,specificity was 76.1% and AUC was 0.850 with 95% confidence interval of 0.776 ~ 0.932.Conclusion: Serum miRNAlet-7a levels in patients with laryngeal cancer are down-regulated compared with the health.miRNALet-7a is expected to be a new molecular marker for the diagnosis of laryngeal cancer;There is a certain correlation between serum miRNAlet-7a level and miRNAlet-7a in tumor tissues.The change of serum miRNAlet-7a level is related to lymph node metastasis,which is helpful for tumor disease judgment and postoperative disease monitoring,but further research is needed.
