Objectives: To respectively detect the expressions of S100 calcium binding protein A7 and Nuclear factor kappa B p65 in normal cervical tissues,cervical intraepithelial neoplasia(CIN)tissues and cervical squamous cell carcinoma tissues while finding the expression difference and correlation among these groups.From theses analyses the relationship between the expressions of S100A7 and NF-κBp65and the clinical or pathological indicators of patients with cervical squamous cell carcinoma was quantified,followed by exploring the relations of their expressions in cervical squamous cell carcinoma to the occurrence,invasion and metastasis of tumors.Methods: A total of 172 cervical specimens from patients who had received excision for hysteromyoma,cervical intraepithelial neoplasias or cervical squamous cell carcinoma in Department of Gynaecology,the Affiliated Hospital of Qingdao University from May 2008 to April 2013 were selected.Expressions of S100A7 and NF-κBp65 in 27 cases of normal cervical tissues,65 of cervical intraepithelial neoplasias tissues(including 28 lower-level cervical intraepithelial neoplasia and 37high-level ones)and 80 of cervical squamous cell carcinoma tissues were detected using Immunohistochemistry(IHC),and semi-quantitative analyzed with software Aperio Image Scope to find the differences and correlations.With collected and arranged clinical and pathological information of patients with cervical squamous cell carcinoma as well as their living status during the follow-up survey after five years of surgery,it was qualified using statistical methods the relations of their expression levels with each clinical and pathological indicators,prospectively showing the expression correlation of them in cervical squamous cell carcinoma.Results: 1.Both the S100A7 and NF-κB p65 were found for their expressions in normal cervical tissue,lower-level and high-level cervical intraepithelial neoplasia tissues along with cervical squamous cell carcinoma tissues.2.The expressions of S100A7 in normal cervical tissue,lower-level and high-level cervical intraepithelial neoplasia tissues were on the increase with differences between any two groups statistically significant(P<0.05).Expressions of S100A7 in cervical squamous cell carcinoma tissues were significantly higher than that in normal cervical tissues orlower-level cervical intraepithelial neoplasia tissues,and were significantly lower than that in high-level cervical intraepithelial neoplasia tissues,with the differences statistically significant for both(P<0.05).The NF-κB p65 showed also increasing expressions in normal cervical tissue,lower-level and high-level cervical intraepithelial neoplasia tissues as well as cervical squamous cell carcinoma tissues.The differences of any two groups were statistically significant(P<0.05).Expressions of S100A7 and NF-κB p65 were correlated with the degree of differentiation of tumor,FIGO stages,lymphatic metastasis and living status after five years of surgery.It was not found a correlation of their expressions with age of patients,tumor size or deep interstitial infiltration.Unlike NF-κB p65,S100A7 had its expression nothing to do with vascular metastasis or para-uterine metastasis.3.There was a positive correlation between S100A7 and NF-κB p65 for their expressions in cervical squamous cell carcinoma.Conclusions: 1.S100A7 and NF-κB p65 have higher expressions in cervical squamous cell carcinoma and high-level intraepithelial neoplasia than in normal tissues or low-level intraepithelial neoplasia.Their expressions were related to the differentiation of tumor,FIGO stages,lymphatic metastasis and living status after five years of surgery.The increase of their expressions involved the occurrence,progression,infiltration and metastasis of cervical squamous cell carcinoma.2.There was a positive correlation between S100A7 and NF-κB p65 expression in cervical squamous cell carcinoma,The two may play a synergistic role in the occurrence,invasion and metastasis of cervical squamous cell carcinoma.
