Objective:E3 ubiquitin ligase UHRF2 is involved in the process of cell proliferation and differentiation,and plays an important role in cell cycle mechanism,regulation of ubiquitin-proteasome system and epigenetic system.Its absence affects the process of normal differentiation of these cells,which may lead to the occurrence of human tumors and certain diseases.It has been shown in many studies that UHRF2 is not expressed in many kinds of tumors and may be a new direction for the diagnosis and treatment of human tumors.However,the study of E3 ubiquitin ligase UHRF2 in prostate cancer has not been reported.In this paper,the expression of UHRF2 in prostate cancer and its clinical significance were discussed in order to find a new and more valuable way for the diagnosis and treatment of prostate cancer.Methods:1.Detection of UHRF2 expression in 78 cases of prostate cancer tissues and paracancerous tissues by immunohistochemical method,Analysis of the relationship between UHRF2 expression in different tissues and clinicopathological characteristics of patients with prostate cancer.2.Detection of UHRF2 protein expression in prostate cancer tissues and paracancerous hyperplasia tissues of 40 patients with prostate cancer by Western blot.Results:1.The results of immunohistochemistry showed that in 78 cases of prostate cancer,17.9%(14/78)had high expression of UHRF2,and 35.9%(28/78)had high expression of UHRF2 in benign prostatic tissue of paracancerous hyperplasia.Compared with the adjacent normal tissues,the expression of UHRF2 was down-regulated in prostate cancer tissues.The difference of UHRF2 was statistically significant in patients with different Gleason scores(P = 0.036),PSA(P = 0.038),TNM stage,P = 0.018),but there was no significant difference in patients with different age(P = 0.555)and lymph node metastasis(P= 0.570).2.Western blot results showed that the expression level of UHRF2 protein in paracancerous hyperplasia tissues was significantly higher than that in paracancerous hyperplasia tissues(P < 0.05).Conclusion:Compared with the adjacent tissues,the expression of UHRF2 was down-regulated,and significantly correlated with Gleason score,PSA,TNM stage.Itsdeletion may be involved in the carcinogenesis and development of prostate cancer,and plays a role as a tumor suppressor.It can be used as an important target for clinical diagnosis and treatment of prostate cancer.
