Prostate cancer is a common malignant tumor in male genitourinary system, it morbidity is increasing in recent years. Some related studies show that it's tumorgenesis and biological characters are closely related to many tumor suppress genes. PTEN ( Phosphatase and Tensin homolog deleted on chromosome TEN) , also named MMAC 1 /TEP 1, which was found in 1 997,has attracted highly attention and been undertaken wide studies. To study the expression and significance of tumor suppressor gene PTEN in prostate cancer, We employed Streptavidin Biotin peroxidase Complex (SABC) methods, Monoclonal antibody against PTFN protein on archival prostate cancer materials. Then use cationic liposome LipofectAmine2000 to delivery retrovirus vector PBabe-PTEN-PURO which contains PTEN and PBabe-PURQ into PTEN-deficient prostate cancer cell line PC-3 respectively to study cell growth, adhesion, segregation, apoptosis, cell cycle, ultrastructure alteration in Vitro and nude mice tumorgenesis ability in Vivo. The results are as follows: PTEN protein was detected in 21 out of 38 cases of prostate cancer, and in 24 cases of well and moderately differentiated prostate cancer positive rate was higher than that of 14 cases of poorly differentiated prostate cancer (P
