Objective: To investigate whether Shexiang Baoxin Pill protects oxidative damage of H9c2 cardiomyocytes induced by high glucose produced by the Nrf2-ARE signaling pathway.Improve the specific mechanism of Shexiang Baoxin Pill in the treatment of diabetic cardiomyopathy.Method: A high-glucose model was established by using high glucose(35mmol/L)treat H9c2 cardiac cells for 24 h.Then,H9c2 cardiac cells were randomly divided into different group,different treatment factors deal with cardiac cells separately.After 24 h,MMT was used to record each experimental treatment group's OD value and measure cell viability;Honest 33258 was used to detect apoptosis of each treatment group;Detection of reactive oxygen species levels in cells of each treatment group by experimental reactive oxygen species(ROS)kit;Nrf2,HO-1 levels were determined by Western blot.Result:(1)Compared with the blank control group,three groups of H9c2 cardiomyocytes were treated in high glucose environment(35mmol/L)for 24 h,cell proliferation decreased,apoptosis was significantly increased(P<0.05);Compared with the high glucose injury group,Shexiang Baoxin Pill could inhibit the apoptosis of H9c2 cardiomyocytes and promote proliferation.The cell proliferation was more significant after treatment with 2g/L of heart pill for 24h(P<0.05).(2)compared with the normal control group,after treatment with high glucose(35mmol/L)for 24 h,the changes of the nucleus were observed under the fluorescence microscope.The results showed that the nucleus of the normal control group was normal.The nucleus has the same size and uniform morphology,and the nucleus is uniform.The chromatin of the high glucose injury group showed pyknosis,and the nucleus was densely stained and nuclear division.After the intervention with shexiang Baoxin Pill,with the increase of drug concentration(1g/L,2g/L),Nuclear division appeared to be significantly reduced(P<0.05);(3)compared with the blank control group,the fluorescence value of H9c2 cardiomyocytes increased and the ROS level increased significantly in the high glucose injury group.After treatment with Shexiang Baoxin Pill,the level of ROS was decreased in a concentration-dependent manner,and the concentration of scented scented sputum was 2 g/L(P<0.01);(4)compared with the high glucose injury group,the high glucose + Shexiang Baoxin Pill(2g/L)group can significantly increase the protein expression level of Nrf2 and HO-1 in cardiomyocytes(P<0.01).Conclusion: Shexiang Baoxin Pill reduces H9c2 cardiomyocyte apoptosis and oxidative stress in high glucose environment by activating Nrf2-ARE pathway.
