Objective Gastric cancer is one of the most common tumors.The incidence and mortality of gastric cancer in China are particularly high.Therefore,the study about mechanism of gastric cancer is particularly urgent.Tribbles homolog 3 is a kind of pseudo-protein kinases,which is involved in the regulation of multiple signaling pathways in human,and mediates the development of various diseases,such as diabetes,coronary heart disease,obesity,Alzheimer's disease.Researches has revealed that TRIB3 is found to be highly expressed in a variety of tumors and leads to the tumor proliferation,invasion and metastasis.However,few reseach studies the influence of TRIB3 on gastric cancer.We aim at figuring out the role of TRIB3 in gastric cancer.It is proposed to define the interacting proteins of TRIB3 in gastric cancer cells,the signaling pathways involved in regulation and the main functions,and to explore the relationship between TRIB3 and clinical and pathological features of gastric cancer patients.Materials and Methods 1.Bioinformatics analysis: There are 415 gastric cancer specimens in the Link Omics online database,these are all from the TCGA database.Each specimen has complete clinical pathological data.Bioinformatics analysis was used to explore the related proteins of TRIB3 in gastric cancer cells,GO analysis and KEGG analysis were performed.,too.Finally,the correlation between TRIB3 and clinical data of gastric cancer was analyzed.2.Immunohistochemistry: This study collected 60 specimens of gastric adenocarcinoma and corresponding specimens of adjacent tissues.The specimens were all from the affiliated hospital of Qingdao University.After screening the regional targets,tissue chip wax blocks were fabricated.The tissue chip was sliced.The expression of TRIB3 in gastric cancer cells and adjacent tissues was detected by immunohistochemistry.Results 1.Results of biosignal analysis:(1)In gastric cancer cells,TRIB3 had 12386 related proteins(P<0.05).There were 5987 genes with positive correlation,such as C20orf20(p=3.21-E42),UBE2C(p=7.62-E41);6399 genes with negative correlation,such as PDE4D(p=7.73E-34),FAM13B(p=1.61E-32).(2)GO analysis was used to clarify the function of TRIB3 in gastric cancer cells,the results showed that TRIB3 has a positive correlation with NF-k B binding function and t RNA binding function,and has a significant negative correlation with ?-catenin binding function and cytokine binding function..(3)Using KEGG analysis to explore the signaling pathway regulated by TRIB3 in gastric cancer cells,we found that TRIB3 is closely related to the resection and repair of bases,cell cycle and steroid biosynthesis.(4)The higher the expression of TRIB3,the higher the partial stage(T stage).The older the gastric cancer patients,the higher the expression of TRIB3.The expression of TRIB3 was also different in different races.The expression of TRIB3 in black race was the highest in the pathogenesis of gastric cancer,followed by yellow race and white race.2.Results of immunohistochemistry:(1)The positive rate of TRIB3 in 60 cases of gastric cancer specimens was 46.7%(28/60),while the positive expression rate of TRIB3 in adjacent tissues was 20%(12/60).The expression of TRIB3 in cancer tissues was higher than that in adjacent tissues(P=0.002).(2)Analysis of the expression of TRIB3 and clinical pathological factors: The expression of TRIB3 was closely related to the TNM stage and distant metastasis of gastric cancer(P<0.05),but there was no significant correlation with age,gender,tumor size and differentiation degree(P>0.05).Conclusion Bioinformatics analysis found that TRIB3 has multiple related proteins in gastric cancer cells and participates in the regulation of multiple signaling pathways.The higher the T stage,the higher the expression of TRIB3.We performed immunohistochemistry and found that TRIB3 is associated with TNM staging and distant metastasis of gastric cancer,while TNM staging guides patients' treatment strategies to predict prognosis and survival time.TRIB3 may be an effective indicator for evaluating the biological behavior of gastric cancer,or it may guide the treatment and has predictive effect of gastric cancer.
