Objective: Researching the expression of CD44v6 and FOXD3 in gastric cancer tissue and adjacent normal tissue, analyzing the relationship between two of them and clinical pathological parameters, exploring relationship between two protein and biological behavior(infiltration, metastasis and prognosis)in gastric carcinoma. It studies the possibility of taking FOXD3 as the marker of gastric cancer stem cell by analyzing the expression of CD44v6 in gastric cancer tissue and the statistical analysis of correlation of FOXD3 and CD44v6. Methods: The first part: selecting 100 surgical specimens of gastric adenocarcinoma, among them there are 11 cases of well differentiated adenocarcinoma, 38 cases of moderately differentiated adenocarcinoma,51 cases of poorly differentiated adenocarcinoma( including mucous cell carcinoma, mucinous adenocarcinoma) and 40 normal gastric mucosa. Using immunohistochemistry method(Envision two step method) to detect the expression of CD44v6 and FOXD3. Analyzing their relationship between clinical pathological parameters in patients with gastric cancer and analyzing the correlation between them. The second part: Taking 125 cases of the department of pathology in University of Virginia from 1982 to 2002, and 60 gastric cancer cases in Pathology Department of No. 1 Affiliated Hospital of Anhui Medical University. There are 185 cases in total. The above two groups of specimens of gastric cancer, respectively, made two gastric cancer tissue microarray combined with immunohistochemistry method(Envision two step method) to detect the expression of CD44v6 and FOXD3, analysis with both the relationship between clinical pathological parameters in patients with gastric cancer and the correlation of the expression in gastric carcinoma, and compared with the results of the first part. Results: The first part:(1) In the gastric cancer tissue and normal tissue adjacent to carcinoma, the detection of expression rate of CD44v6 is respectively 88%(88/100)and 22.5%(9/40). The differences of the two has statistical significance(χ2=57.601,P<0.001). The detection of the expression of CD44v6 has something to do with differentiated degree, the depth of infiltration, the transfer of lymph gland and clinical stages(P<0.05). It has nothing to do with the gender, age and the size of tumor(P>0.05).(2) In the gastric cancer tissue and normal tissue adjacent to carcinoma, the detection of the expression rate of FOXD3 is respectively 22%(22/100) and 90%(36/40). The differences between them has statistical significance(χ2=49.694,P<0.001).The detection of the expression of FOXD3 has something to do with differentiated degree, the depth of infiltration, the transfer of lymph gland and clinical stages(P<0.05).It has nothing to do with the age, gender and the size of tumor(P>0.05).(3)In the expression of gastric cancer tissue, CD44v6 and FOXD3 was significantly negatively related(rs =-0.276,P<0.01). The second part:(1) n the gastric cancer tissue and normal tissue adjacent to carcinoma, the positive expression rate of CD44v6 is 47.27%(52/110) and 22.5%(9/24), and it statistical significance with the first positive expression rate differences between normal tissue adjacent to carcinoma(χ2=7.461,P<0.01).Positive expression of CD44v6 protein associated with tumor invasion depth, the transfer of lymph gland and clinical stages(P<0.05), It has nothing to do with the patient’s age, gender, the size of tumor, and the degree of differentiation(P>0.05), consistent with the first part of the results except the expression of differentiation;(2) In gastric cancer tissue and normal tissue adjacent to carcinoma, FOXD3 protein positive expression rate of 30.91%(34/110) and 90%(36/40), with the first part of the normal tissue adjacent to carcinoma positive expression rate differences statistically significant(χ2=36.747,P<0.001); FOXD3 protein positive expression is associated with tumor invasion depth, the transfer of lymph gland and clinical stages(P<0.05), It has nothing to do with the patient’s gender, age, the size of tumor and the degree of differentiation(P>0.05), consistent with the first part of the results except the expression of differentiation;(3) in gastric cancer tissue, the expression of CD44v6 protein and FOXD3 protein was significantly negatively related(rs =-0.583,P<0.001). Conclusion:(1) As the tumor marker of gastric cancer stem cell, CD44v6 is closely related with the biological behavior(infiltration, metastasis and prognosis)in gastric cancer.(2) As the key transfer factor of stem cell, FOXD3 is closely related with the biological behavior(infiltration, metastasis and prognosis)in gastric cancer.(3)FOXD3 and expression of CD44v6 protein is significantly negatively related. They may restrict each other with the invasion and metastasis of gastric cancer. FOXD3 may be the tumor marker of gastric cancer stem cell.(4) Tissue chip is the Clinical tissue samples of high throughput and high efficiency. It is more quick, economical, time-saving and labor-saving than traditional section preparation.
