Objective:To study the changes of serum and tissue Irisin in breast cancer patients and the correlation between clinicopathological features,and to explore the expression level of Irisin in breast cancer patients and its clinical significance.Methods:Blood samples were collected from 46 patients with breast invasive carcinoma,30 patients with breast fibroadenomas,and 12 healthy subjects who underwent physical examination in our hospital.The expression of Irisin in three groups of fasting serum was detected by ELISA.Level;Surgical resection and pathologically confirmed breast fibroadenoma tissue in 20 cases,invasive breast cancer tissue in 20 cases,and selected corresponding adjacent tissues(at least 5cm from the edge of the cancer tissue)were collected from the breast cancer clinic of the Affiliated Hospital of Qingdao University.The expression level of FRNC5 of Irisin precursor in the three groups was detected by Western blot.Statistical methods were used to analyze the expression level of Irisin in breast cancer patients with different body mass index(BMI),and to analyze the correlation between the expression level of Irisin in serum and tissues and the clinicopathological features of breast cancer.Results:The serum levels of Irisin in gland invasive carcinoma group,breast fibroadenoma group and healthy person group were(2.15±0.53),(3.08±0.44),(3.26±0.34),respectively.The serum Irisin level of breast cancer patients was significantly lower than that of breast fiber.Adenoma group and healthy person group(t=8.03,6.93,P<0.01);Serum Irisin levels in the breast fibroadenoma group were not significantly different from those in the healthy group(P>0.05).Serum Irisin expression in lymph node metastatic breast cancer patients was significantly lower than that in patients without lymph node metastasis(t=2.38,P<0.05);The expression level of serum Irisin in Ki67>20% breast cancer patients was significantly lower than that in Ki67?20%(t=2.89,P<0.05).Western blot analysis showed that the relative expression of Irisin protein in breast cancer group,breast cancer side group and benigngroup were(0.906±0.556),(1.166±0.649),(1.548±0.624),and Irisin in cancer tissues.The relative expression of protein was significantly lower than that of the paracancerous group and the benign group(t=-3.045,7.684,P<0.05),and the difference was statistically significant.The paracancerous group was compared with the benign group(t=4.244,P<0.05),and the difference was statistically significant.Correlation between the levels of irisin in serum and body mass index(BMI)in breast cancer patients showed a positive correlation between iris and BMI,P < 0.05,the difference was statistically significant.Conclusion:The expression level of serum Irisin is closely related to breast cancer proliferation and lymph node metastasis.The expression level of Irisin has a certain correlation with the clinicopathological features of breast cancer.Detection of Irisin may provide a reference for understanding the mechanism of breast cancer development and prognosis.
