Objectives To investigate the expression of DCC and MKK4 in human colorectal cancer and its relationship with clinicopathological factors.MethodsCollect the cancer tissues and adjacent tissues of 60 patients with colorectal cancer diagnosed in the Affiliated Hospital of North China University of Science and Technology from September 2017 to August 2018 with complete clinical data.Among them,38 were males and 22 were females;36 were younger than 60 years old and 24 were younger than 60 years old;32 were tumors located in the colon and 28 in the rectum;25were protuberant,30 ulcerative and 5 infiltrative;12 were highly differentiated,40 were moderately differentiated and 8 were poorly differentiated;the depth of infiltration was T14,T2 14,T3 36 and T4 6;41 were not accompanied by lymphatic metastasis,19 with lymphatic metastasis;50 cases without distant metastasis and 10 cases with distant metastasis.Immunohistochemistry was used to detect the expression of DCC and MKK4in colorectal cancer tissues and adjacent tissues.Realtime PCR was used to detect the expression of DCC and MKK4 in colorectal cancer tissues and adjacent tissues.Western blotwas used to detect the expression of DCC and MKK4 in colorectal cancer tissues and adjacent tissues.To analyze the correlation between DCC and MKK4 expression.To analyze the relationship between the expression of DCC and MKK4 and clinicopath-ological factors in patients with colorectal cancer.ResultsImmunohistochemical results showed that the positive rate of DCC protein expression was 20.0%in colorectal cancer tissues was 96.7%in adjacent tissues,with significant difference?P<0.05?;the positive rate of MKK4 protein expression was 39.5%in colorectal cancer tissues was 95.5%in adjacent tissues,with significant difference?P<0.05?.Realtime PCR results showed that the relative expression of DCC in colorectal cancer tissues was?0.361±0.078?,lower than that in adjacent tissues?1.000±0.000?,with significant difference?t=14.074,P<0.001?;the relative expression of MKK4 in colorectal cancer tissues was?0.442±0.045?,lower than that in adjacent tissues?1.000±0.000?,with significant difference?t=21.371,P<0.001?.Western blot results showed that the relative expression of DCC protein in colorectal cancer tissues was?0.184±0.073?,lower than that in adjacent tissues?0.995±0.078?,with significant difference?t=13.147,P<0.001?;the relative expression of MKK4 protein in colorectal cancer tissues was?0.158±0.081?,lower than that in adjacent tissues?1.020±0.051?;the difference was statistically significant?t=15.57,P<0.001?.There was a positive correlation between the expression of DCC and MKK4 in colorectal cancer?r=0.628,P<0.05?.The expression of DCC and MKK4 protein was significantly correlated with the degree of differentiation,lymphatic metastasis and distant metastasis?P<0.05?,but not with gender,age,tumor location,gross morphology and depth of invasion?P>0.05?.Conclusions 1.DCC and MKK4 were low expressed in colorectal cancer.2.The expression of DCC and MKK4 in colorectal cancer is positive correlated.3.The expression levels of DCC and MKK4 are related to the degree of differentiation,lymphatic metastasis and distant metastasis of colorectal cancer.Figure 4;Table 10;Reference 140...
