Objective: To investigate the effects of Rho-kinase(ROCK)inhibitor RKI-1447 on cell proliferation,migration,extracellular matrix synthesis in rat urethral fibroblasts induced by TGF-?1,as well as the expression of tissue inhibitor of metallo-proteinase 1(TIMP-1)and cytoskeleton-associated proteins,provid theory basis for prevention and treatment of urethral stricture in clinic.Methods : Fibroblasts derived from rat urethra were cultured and identified by anti-vimentin immunocytochemistry.The fibroblasts were divided into three groups.Except for no intervention for normal group,other groups were incubated with TGF-?1(10?g/L)and RKI-1447 in different concentrations(0,1,2,4,8,16?mol/L).The cell proliferation was evaluated by MTT assay.The cell migration ability was detected by Transwell assay.The expression of ROCK-1,collagen I,collagen III,TIMP-1,?-SMA and p-MLC was detected by Western-blot analysis.Results : TGF-?1 could promote the proliferation and migration of rat urethral fibroblasts and induce the expression of ROCK-1,collagen I,collagen III,TIMP-1,?-SMA and p-MLC.Compared with control group,RKI-1447 could inhibit the proliferation and migration of rat urethral fibroblasts.With increasing concentration of RKI-1447 intervention,the expression of ROCK-1,collagen I,collagen III,TIMP-1,?-SMA and p-MLC decreased gradually.Conclusion:RKI-1447 could inhibit the proliferation,migration,collagen production and expression of TIMP-1,?-SMA and p-MLC of rat urethral fibroblasts induced by TGF-?1 in vitro.Further studies are deserved to investigate the effects of RKI-1447 on the prevention and treatment of urethral stricture in clinic. |
