Effect And Possible Mechanism Of Celecoxib On The Formation Of Scar After Internal Urethrotomy In Rat

Objectives: This study evaluated the efficacy of celecoxib in the prevention of formation of scar after internal urethrotomy in rat and explored the possible mechanism.Methods: Thirty-two male Wistar rats were randomly divided into three groups:Group A,Group B,Group C.Group A were the control group.Group B underwent internal urethrotomy.Group C were done the same operations and administration of Celecoxib (20mg/kg) everyday.Rats in each group were sacrificed after 12 weeks.The surface density was used to evaluate the degree of urethral collagen fiber formation.The expressions of TGF-β1 mRNA and Smad7 mRNA were measured by real-time fluorescence quantitative PCR.Results: 1.The surface densities of urethral collagen fibers in Group A, GroupB, Group C were (19.86±5.88)%,(38.63±7.43)%and (28.14±5.96)% respectively. There were significant differences in three groups (F=76.27,P<0.05).Compared with Group A, the surface densities of GroupB and Group C increased. Compared with Group B, the surface densities of GroupB and Group C decreased. 2.The gene expressions of TGF-β1 at mRNA level in Group A, GroupB, Group C were 0.322±0.129,0.708±0.103,0.542±0.127 respectively. There were significant differences in three groups (F=31.11,P<0.05).Compared with Group A, the gene expressions of TGF-β1 mRNA of GroupB and Group C increased. Compared with Group B, the gene expression of TGF-β1 mRNA of Group C decreased.The gene expressions of Smad7 at mRNA level in Group A, GroupB, Group C were 0.422±0.059 , 0.172±0.073 , 0.258±0.067 respectively. There were significant differences in three groups (F=43.67,P<0.05)).Compared with Group A, the gene expressions of Smad7 mRNA of GroupB and Group C decreased. Compared with Group B, the gene expressions of Smad7 mRNA of Group C increased.Conclusions:1. The gene expressions of TGF-β1 mRNA Smad7 mRNA are detected in all groups.2.
