ObjectiveTo investigate the role of Protein O-linked mannose ?1,2-N-acetylglucosaminyltransferase 1(POMGnT1)in the pathogenic process of Alzheimer's diseaseMethodsThe expression of POMGnT1 gene was examined in AD model cells(N2a/APP695swe)and N2a/wt cells using Real-time PCR and western blot This expression was also examined in AD model mice(APP/PSEN1)and wild-type mice using immunofluorescence staining.A? deposits were examined in POMGnTl-gene-knockout mice and wild-type mice using immunochemi stry.ResultsWe found that the expression of POMGnTl gene decreased in mRNA(0.80±0.02)and protein(0.50±0.019)levels in N2a/APP695swe cells than N2a/wt cells(mRNA:1;protein:1.31±0.039;both P<0.01)Immunofluorescence results showed the reduced expression of POMGnT1 protein in neurons of APP-PSEN1 mice.Immunochemistry results showed more A? deposits in POMGnTl-gene-knockout mice(2m:wild type,0.048±0.001 vs.POMGnT1-/-,0.358 ± 0.014,P<0.01;1y?wild type,0.229 ±0.0025 vs.POMGnT1-/-,0.266±0.0041,P=0.0015)ConclusionThese findings suggest POMGnT1 gene may play an important role in the pathogenic process of Alzheimer's disease.
