Aims: 1. To determine the relationship of three SNPs(R702W, G908R and 3020insC) of CARD15gene with the susceptibility to Crohn's Disease in Chinese population in Suzhou Area.2.To investigate the role of CARD15 gene in the process of signaling, and the changes of O-glycosylation in the development of Crohn's disease.Methods:1. Twenty Chinese Crohn's disease patients had genotyping performed using AS-PCP and PCR-RFLP against the wild-type and the R702W, G908R and 3020insC variants of the CARD15 gene. Controls consisted of 20 ulcerative colitis patients and 20 healthy persons. 2. We used reverse-transcription polymerase chain reaction(RT-PCR) to detect the mRNA expression of CARD15 gene and ppGalNAc-T family in CD, UC and healthy person, andβ-actin was used as control..Resμlts:1. None of the Crohn's disease patients had heterozygous and homozygous SNP variants. None of the ulcerative colitis and healthy controls had these SNPs. 2. The mRNA expression of CARD15 in CD is higher than UC and healthy person. The expression of ppGalNAcT1,T4 and T7 was sharply dropped, T11 expressed more, and T12 appeared to express.
