Objective:1.Detect the expression level of macrophage colony stimulating factor(M-CSF)and tumor necrosis factor-?(TNF-?)in pre-eclampsia(PE)placenta.2.Explore the effects of M-CSF and TNF-?on the invasion and excessive inflammation of placental trophoblast cells,which is helpful to further understand the pathophysiological mechanism of PE.Methods:From September 2015 to September 2017,77 cases of PE were collected from the Department of Pathology of the First Affiliated Hospital of Henan University of Science and Technology.The placental tissue was normal and the normal group(including 35 cases of early-onset PE group and 42 cases of late-onset PE group).Thirty pregnant women(including 10 cases?34 weeks,20 cases>34 weeks)were the control group.Immunohistochemical SP method was used to detect the expression of M-CSF and TNF-?in placenta of each group,and the correlation between them was analyzed.Results:1.The expression of M-CSF and TNF-?in PE placenta was significantly higher than that in normal placenta,and the difference was statistically significant(?~2=18.59,P<0.05;?~2=30.21,P<0.05).2.The positive expression rates of M-CSF in the control group,late-onset PE group and early-type PE placenta were 30%(9/30),61.90%(26/42),and 82.86%(29/35),respectively;M-CSF The positive expression in the early-onset PE placenta tissue was significantly higher than that in the control group(?~2=18.59,P<0.05),and the difference was statistically significant.The expression of M-CSF in the late-type PE placenta tissue was significantly higher than that of the control group(?~2=7.13).P<0.05),the difference was statistically significant.The positive expression of M-CSF in early-onset PE placenta tissue was higher than that in late-type PE group(?~2=4.02,P<0.05).3.The positive expression rates of TNF-?in the control group,late-onset PE group and early-type PE group were 33.33%(10/30),69.05%(29/42),and 91.43%(32/35),respectively;TNF-?The positive expression in the early-onset PE placenta tissue was significantly higher than that in the control group(?~2=21.37,P<0.05),and the difference was statistically significant.The expression of TNF-?in the late-type PE placenta tissue was significantly higher than that of the control group(?~2=8.99,P<0.05),the difference was statistically significant;The positive expression of early-stage PE placenta tissue was higher than that of late-type PE group,the difference was statistically significant(?~2=4.53,P<0.05).4.In PE,Spearman rank correlation analysis revealed that there was a significant positive correlation between TNF-?and M-CSF expression in PE placenta(?_s=0.546,P<0.05).Conclusions:1.M-CSF and TNF-?may play a role in the pathogenesis of PE.2.The positive expression of M-CSF and TNF-?in placental tissue increased with the severity of PE.3.The expression levels of M-CSF and TNF-?in PE placenta were positively correlated,suggesting that they may play a synergistic role in the development of PE,providing new reference and ideas for the diagnosis and treatment of PE.
