Objective:Colon cancer(CC)is a common malignant tumor of the digestive tract.China's 2015 cancer statistics show that China's colorectal cancer morbidity and mortality ranks in the top five among all malignant tumors,including 376,000 new cases and 191,000 deaths.The high incidence and high mortality of colon cancer are associated with the malignant invasive pathological features of colon cancer.Because the overall evaluation of colon cancer is not high,some patients with colon cancer fail to standardize treatment,and the prognosis is poor.There are still no specific molecular markers that can be used to assess the malignancy,treatment,and prognosis of a tumor.miRNAs are endogenous,non-coding small RNAs of approximately 19 to 25 nucleotides in length.It is an important regulatory molecule for the expression of functional genes and proteins in human cells.By interacting with the 3' untranslated regions(UTRs)of the target mRNA,or by scission and degradation of the target mRNA by silencing the complex,regulation of gene expression and translation of the protein at the post-transcriptional level,thereby affecting the cell's biology Learning function.There are many abnormally expressed miRNAs in colon cancer tissues.Different abnormally expressed miRNAs play different roles in colon cancer,and their application value in clinical practice is different.Subsequent studies have shown that the functional characteristics of miRNAs are mainly dependent on the target gene.A miRNA can regulate multiple target genes,and multiple miRNAs may act on the same target gene,forming a complex regulatory network in the cell.However,different miRNAs are located differently in the regulatory network.The key core molecules in the regulatory network can affect the biological characteristics of tumor cell proliferation,migration and invasion through various target genes,which can affect the clinicopathological features of the tumor,such as tumor size,differentiation degree,lymph node metastasis,distant metastasis.In recent years,the role of miRNA-135b-5p in tumors has received increasing attention from researchers.miRNA-135b-5p plays an important role in a variety of solid tumors,and participates in the formation of tumors as an oncogene or tumor suppressor gene,and plays an important role in the biological characteristics such as proliferation and invasion of tumor cells.In this study,real-time quantitative PCR was used to detect the expression level of miRNA-135b-5p in colon cancer tissues and paracancerous tissues,and the relationship between miRNA-135b-5p expression level and clinicopathological features of colon cancer was analyzed.-5p provides a basis for assessing the malignant degree of tumor and determining the molecular target of prognosis,and further provides candidate genes and possible therapeutic targets for functional research of miRNA.Method:1.Case collection:Inclusion criteria(1)From September 2017 to October 2018,colonoscopy and postoperative pathological examination in the endoscopy room of the Affiliated Hospital of Chengde Medical College were diagnosed as colon cancer;(2)were all found colon cancer for the first time.Any chemotherapy,radiotherapy and targeted therapy was not using;(3)the patient's clinical pathology data is complete.Exclusion criteria(1)patients with a family history of hereditary intestinal diseases(familial adenomatous polyposis,hereditary nonpolyposis colon cancer);(2)patients with other sites of tumor or partial colon resection;3)patients after cholecystectomy;(4)patients with colon adenoma,inflammatory bowel disease.A total of 42 cases of colon cancer were included,and 42 cases of paracancerous mucosa were paired.There are 28 males and 14 females.The age ranged from 48 to 82 years,with an average age of 64.95 ± 8.34 years.2.According to the(AJCC)/(UJCC)colon cancer TNM staging system(7th edition),in which the tumor invades the submucosa as T1,the intrinsic muscular layer is T2,and the tumor penetrates the muscularis propria The paracolic tissue reaching the subserosal layer or invading the non-peritoneal covering is T3,and the tumor penetrating the peritoneal visceral layer is T4.There was no regional lymph node metastasis to N0,1-3 regional lymph node metastasis to N1,and more than 4 regional lymph node metastasis to N2.No distant transfer to M0,and distant transfer to M1.According to the 2010 WHO colon adenocarcinoma grading criteria,>95% of duct formation is highly differentiated,50%-95% of duct formation is moderately differentiated,and 0-49% of duct formation is poorly differentiated.The clinicopathological features of 42 colon cancer tissues were collected from the Department of Pathology,Affiliated Hospital of Chengde Medical College,including 21 patients with tumor diameter >5 cm,21 patients with ?5 cm,and the colon with spleen and 33 patients with right colon.9 cases of colon;15 cases of invasive depth T2-T3,27 cases of T4;24 cases of lymphatic metastasis,18 cases of no lymph node metastasis;8 cases of distant metastasis,34 cases without distant metastasis;5 cases of high school differentiation,moderate differentiation 29 cases,8 cases of moderate to poor differentiation.The clinical stage was 12 cases in stage II,22 cases in stage III,and 8 cases in stage IV.All pathological sections were confirmed by the deputy chief physicians of two pathology departments to meet the WHO colon adenocarcinoma criteria.3.Real-time quantitative PCR was used to detect the expression level of miRNA-135b-5p in colon cancer tissues and adjacent mucosa tissues.4.Analyze the data using SPSS 19.0.The normal distribution measurement data is represented by X ±S.The difference of miRNA-135b-5p between colon cancer tissues and paracancerous mucosa tissues and the clinicopathological features of colon cancer were compared by t test.Chi-square test was used to analyze the correlation between miRNA-135b-5p expression and clinicopathological features.P<0.05 was considered statistically significant.Result:1.Comparison of the expression levels of miRNA-135b-5p in colon cancer tissues and adjacent mucosa tissues.The expression level of miRNA-135b-5p in colon cancer tissues(28.15±0.91)was significantly higher than that in adjacent mucosa tissues(17.93±0.76)(P<0.05),and the difference was statistically significant.2.Comparison of the difference in the expression of miRNA-135b-5p in the clinicopathological features of colon cancer tissues.The relative expression of miRNA-135b-5p was detected by real-time quantitative PCR.The results showed that the expression levels of miRNA-135b-5p in tumor tissues of >5 cm and <5 cm were 31.17±1.00 and 25.13±1.21,respectively(P<0.05).In tumor tissues with invasive depths of T2-T3 and T4,the expression levels were 21.48±0.94 and 31.86±0.53,respectively(P<0.05).The expression levels in tumor tissues without lymph node metastasis and lymph node metastasis were 22.76±1.22 and 31.50±0.49,respectively(P<0.05).The expression levels of tumor tissues with distant metastasis and distant metastasis were 27.27±1.05 and 31.89±0.96,respectively(P<0.05).The expression levels of miRNA-135b-5p in patients with clinical stage III and IV were 30.46±0.97 and 31.89±0.96,which were higher than those in stage 21.43±1.13(P<0.05).The expression of miRNA-135b-5p was 28.34±1.02,32.29±1.12,and the high-differentiation was 20.46±1.97(P<0.05).The difference was statistically significant.There was no difference in the expression of miRNA-135b-5p in the left and right colons(P>0.05).3.The correlation between the expression level of miRNA-135b-5p and the clinicopathological features of colon cancer.The average expression of miRNA-135b-5p in 42 colon cancer tissues was 28.15±0.91.According to the clinicopathological features,each group was divided into high expression and low expression groups.Chi-square test was used to analyze the correlation between different expression groups of miRNA-135b-5p and clinicopathological features.The results showed that the expression level of miRNA-135b-5p was different from tumor size,depth of invasion,lymph node metastasis,distant metastasis and differentiation.Correlation(P<0.05),but no significant correlation with tumor location and clinical stage(P>0.05).Conclusion:The expression of miRNA-135b-5p is significantly increased in colon cancer tissues,and plays a role similar to oncogenes.Its abnormally high expression is closely related to the malignant invasive biological behavior of colon cancer,suggesting that it can be used as a potential target for assessing the malignancy,treatment and prognosis of tumors.
