Based on image processing technology,this study collected and analyzed the dynamic change information of coagulation experiments under 40 times microscope,and obtained the cell formation rate and fusion rate of each experimental group,so as to explore the influence of lectin on the changes of blood agglutination law at the microscopic angle.The main work of this paper is as follows:1.Microscopic design of lectin blood agglutination principle.By comparing the existing blood agglutination monitoring methods of lectin,a new microscope-based agglutination monitoring method and coagulation experiment design were proposed from the microscopic point of view,and the dynamic change information of blood agglutination under the microscope was collected at a rate of one image every 180 seconds.2.Blood agglutination dynamic change information processing.Firstly,the Gaussian filter and the top-hat and bottom-hat transformation are used to complete the preprocessing,and then the gray image of the RGB model and the Cr component image in the YCrCb model are extracted proportionally,and then the Canny operator edge detection and morphological processing are performed or calculated.The results of the preliminary segmentation were finally segmented again using the watershed algorithm to accurately obtain the information of blood agglutination dynamics under different conditions.3.Analysis of the dynamic changes of blood agglutination.In the 8% red blood cell multi-gradient lectin experiment,the cell clustering rate was 18% to 25%,the stabilization time was 30 minutes,and the cell fusion rate was about 80% to 91%.In the 5% red blood cell multi-gradient lectin experiment,the cell clustering rate was16%~21%,and it stabilized in 40 minutes.And the cell fusion rate was 71%~82%.In the 2% red blood cell multi-gradient lectin experiment,the cells The grouping rate was 11%~17%,and it stabilized in about 50 minutes.The cell fusion rate was65%~76%.The fusion rate in all experimental groups tended to be stable for 40~50minutes.Further analysis of each experimental group found that: although the cell aggregation is random during the agglutination reaction,the addition of lectin will affect the clustering rate,and the cell clustering rate was normally distributed with thelectin concentration under the same red blood cell volume fraction,and the lectin rate tends to be the highest when the lectin concentration reaches 250 ?g /mL.As the cell volume fraction becomes larger,the clustering rate increases accordingly.At the same time,the greater the concentration of lectin was and the higher the red blood cell volume fraction was,the shorter the agglutination reaction time was and the higher the cell fusion rate was.The experimental results in this paper fully reveal the related characteristics of lectin on blood agglutination,and further provide a new observation and monitoring method for the experiment of lectin on blood action characteristics.
