Background: Hypertension is a disease affected by both environment and gene.Although there were many candidate genes related to the pathogenesis of hypertension,the reports about these genes or SNPs obviously significant were litte.Add Vitmin B1 to anti-hypertension medicine in therapeutic schedule had better controled blood pressure and improved the vascular function.SLC35F3 encodes a thiamine transporter.Study suggested that SLC35F3 gene was a new candidate gene playing a role in hypertension occurrence and blood pressure regulation.Object: The aim of this study was to investigate the association between the single nucleotide polymorphisms(SNPs)in coding region of SLC35F3 with hypertension in a Chinese population.Method: Sanger Sequencing were performed in 93 samples to find SNPs of coding regions and intron-exon boundaries in SLC35F3 gene,followed by SNP genotyping using Taqman-MGB assays in a total of 2632 subjects for case-control study.Thiamine B1 level in plasma was determined by ELISA kit.Results: We found eight genetic variants in aforementioned regions of SLC35F3.We identified rs34032258,a non-synonymous variant,has a minor allele frequency of 3.9% C>G in 93 samples tested.In the following case control study,G carriers of the minor allele of rs34032258 who are obese had an estimated 2.1% lower risk of hypertension(P =0.038).Analysis of covariance showed that homozygosity for the minor allele of the rs34032258 polymorphism was associated with 8 mm Hg higher diastolic blood pressure levels,13 higher BUN and 14 lower Cr(P =0.007;0.012 and 0.029 respectively).No significant difference was found in SBP between genotypes.Furthermore,linear regression analysis showed that this polymorphism was associated with the change of both SBP(P=0.03)and DBP(P=0.01)after adjustment for age,gender and body mass index in hypertensive patients.Conclusions: SLC35F3 polymorphism rs34032258 is associated with high blood pressure and may increase the risk of hypertension.The new hypertension-susceptibility locus may indicate scientific implications for the pathogenesis and new drug target of systemic hypertension.
