The Relation Between EL And Preeclampsia

Objective:To research the differential expression of EL in placenta both in normal and PE women and to investigate the affective mechanism of EL on human trophoblast cell.Methods:We collected 16 pairs of placenta tissues and used Real-Time PCR?Western Blot?Immunohistochemistry to research whether EL changed in PE people.Additionally,the HTR8/SVneo down-expressing EL was got through being transfected with siRNA-EL.We use Flow cytometry test?Cell scratch test?CCK8?Tube formation to investigate the influence of EL on HTR8/SVneo.Compared with normal one,the influence of EL on many functions of HTR8/SVneo such as apoptosis rate,migration,viability,the ability of Tube formation was evaluated.We also used Real-Time PCR to investigate the molecule mechanism of EL decrease.Results:We found that EL was decreased in placenta of PE people.Besides,the biological activity was declined.Not only the expression of MMP9 was decreased,but also we found the S1PR3 expression was down-regulated as the S1PR1 expression increased.Conclusion:The biological activity of HTR8/SVneo was decreased as expression of EL declined,we consider that these changes may participate in the pathophysiology of PE.We also speculated EL declination affected the S1PR-SPHK-eNOS pathway,which may bring about the inhibitory of trophoblast cell implantation.
