Background:Preeclampsia(PE)is a common and pregnancy specific disease,resulting in the important cause of maternal-fetal morbidity and mortality,its etiology and mechanism were complex,but not clear,may be involved in the placenta,fetal and maternal.Studies have shown that disturbed trophoblast invasion and impaired uterine spiral artery remodeling leading to placental ischemia/hypoxia is an important pathological features of preeclampsia.Therefore,preeclampsia is also known as a placenta ischemic disease.AMPK is a key molecular in regulating energy metabolic,plays an important role in a variety of metabolic diseases by regulating glucose uptake and fatty acids metabolism.So we study the metabolomics of preeclampsia,detect relative molecular to explore the pathological mechanism of preeclampsia,provide the information for clinical prediction and treatment.Methods:Normal placenta and PE placenta were collected and HTR8/SVneo trophoblast was used for setting up PE cell model;the metabonomics of trophoblast were analyzed by GC-MS;q-PCR and Western Blotting detect the mRNA and protein level;MDA describe the lipid oxidation level;ACEA real-time cell analysis and transwell perform the migration and invasion of trophblast.Results:The metabolism disturbed in PE placenta(p<0.05,q<0.05);the phosphorylation of AMPK were significantly elevated in PE placenta(N=16,p<0.05);Hypoxia induced hyperactive of AMPK in trophoblast and impaired the migration and invasion.Conclusion:PE placenta is associated with disturbed metabolism;Trophoblast prefer using short chain fatty acids during hypoxia;Hypoxia induced AMPK hyperactivation result in impaired trophoblast migration and invasion.
