Objective: To study the changes of serum triglycerides and apolipoprotein C?,C?,E before and after the high-fat meal in individuals with different fat tolerance.To investigate the possible underlying mechanisms of hypertriglyceridemia.Methods:We recruited volunteers from the outpatient department of endocrinology,Hebei General Hospital.After all volunteers signed written informed consent,physical examination and oral glucose tolerance test(OGTT)were performed.Individuals with normal OGTT were included in the study.After a week of normal diet washout and 8-hours fasting,all subjects were instructed to take a high-fat meal(total calories 1500 kcal,fat,protein and carbohydrate content 60%,20% and 20% respectively)within 10 minutes.The high-fat meals were prepared by a professional nutritionist.Venous blood samples of fasting and 2,4,6 hours after the high-fat meal were taken for detection.Apo C?,C?,E were measured with enzyme-linked immunosorbent(ELISA)method.The subjects were divided into three groups according to their serum TG levels of fasting and 4 hours after high-fat meal:(1)normal fat tolerance(NFT,0h TG<1.7mmol/L and 4h TG<2.5 mmol/L);(2)impaired fat tolerance(IFT,0h TG<1.7mmol/L and 4h TG ? 2.5mmol/L);(3)hypertriglyceridemia(HTG,0h TG?1.7 mmol/L).SPSS 21.0 software was used for statistical analysis.Results:The level of fasting Apo C? and C? in three groups were not statistically different among three groups(P = 0.129,P = 0.964).There were significant difference among three groups in the level of TG(P < 0.001).At fasting and 2,4,6 hours after the high-fat meal,TG level gradually increased with the decrease of fat tolerance.There was statistical significance between NFT group and HTG group at 6 hours after high-fat meal(P = 0.012),and the Apo C? level in HTG was lower than that in NFT;otherwise,the Apo C? level in three groups were not statistically different at the other time points.Compared with NFT group,the level of Apo C? in HTG group was significantly lower at 2 and 6 hours after high-fat meal(P=0.02;P=0.002).The level of Apo C? was negatively correlated with TG level at 6 hours after high-fat meal(r=?0.181,P=0.038).The level of Apo C? was negatively correlated with TG level at 2 and 6 hours after high-fat meal(r=?0.224,P=0.023;r=?0.189,P=0.046).Conclusion:1.With the decrease of fat tolerance,the peak of serum TG level after high-fat meal was delayed and increased.2.The decreased secretion of apolipoprotein C? and C? in HTG was consistent with dysfunction in TG clearing after high-fat meal.
