Part one The expression of the mi RNA-197 in the Tissue of Oral Squamous Cell CarcinomaObjective: To detect the expression of mi RNA-197 in the Oral cancer tissue and normal oral mucosal tissue and to investigate whether mi RNA-195 has influence on OSCC carcinogenesis and lymphatic metastasis.Methods:1 The expression level of miRNA-197 in OSCC and normal oral mucosa were detected by RT-Qpcr.The correlation between the expressio n level in carcinogenesis and cervical lymph node metastasis were studi ed.2 SPSS21.0 software was used to analysis experimental data.The t test was used to compare the differences between the experime-ntal data of each group,and P<0.05 was considered to have the dif-ference of statistics.Result:1 The expression level of miRNA-197 in OSCC and normal oral mucosa were detected by RT-Qpcr.The correlation between the expressio n level in carcinogenesis and cervical lymph node metastasis were studi ed.2 SPSS21.0 software was used to analysis experimental data.The t test was used to compare the differences between the experime-ntal data of each group,and P<0.05 was considered to have the dif-ference of statistics.Brief Summary:The expression level of miRNA-197 in OSCC tis sue were higher than in normal and significantly correlated with lympha tic metastasis,Which suggest that miRNA-197 may as a Oncogene in th e occurrence、development and migration of OSCC.Part two Study of the effect of mi RNA-197 on the biological function of CAL-27 cellsObjective: To investigate the influences of miRNA-197 on proliferation and migration of CAL-27 and to explore the role of miRNA-197 in carcinogenesis and lymphatic metastasis of OSCC.Methods:1 The recovery,cultivation,passage and frozen storage of OSCC-15 CAL-27 cells.2 Synthetic mi RNA-197 inhibitor and mi RNA-195 NC were transfected into CAL-27,seting experient group、negative control group and blank control group.3 MTS colorimetric analysis was used to detect the OD of 0hour,24 hour,48hour,72 hour,96hour and plot the cell growth curve.4 We transfected the miRNA-197 inhibitor into OSCC cell line,and observed the cloning ability of OSCC cells.5 Transwell assay was used to detect the effect of miRNA-197 on migration of CAL-27 cell.6 The data were analysised by SPSS21.0 software and were expressed as mean ± standard deviation.The t-test was used for paired samples and Mann-Whitney test was used for independent samples between the two groups.Results:1 At 24 hours later,The expressions of miRNA-197 in experient gr oup、negative control group and blank control group were 12.65±0.24、14.85±0.07、14.51±0.08,there were differences(P <0.05).2 After the MTS 48 hours,experient group、negative control group and blank control group’s OD were 0.44±0.04、0.56±0.03、0.58±0.01,t here were differences(P <0.05).3 In the cell clonality experient,experient group、negative control group and blank control group’s formate rate were 3.97±0.26%、7.50±0.42%和 7.78±0.21%,there were differences(P <0.05).4 In the cell clonality experient,experient group、negative control group and blank control group’s formate rate were 3.97±0.26%、7.50±0.42%和 7.78±0.21%,there were differences(P <0.05).5 In the cell transwell experient,the breaking through cell number were 115.33±3.68、163.67±4.01 和 167.33±5.3,there were differences(P <0.05).Brief Summary:Mi RNA-197 is a tumor suppressor in OSCC,which could inhibit the proliferation and migration of CAL-27 cells.Mi RNA-197 might be a new potential target for OSCC therapy.Conclusion: Mi RNA-197 expression level is up-regulated in OSCC tissue,and it is closely associated with the transfer and prognosis of the cancer.The chang of miRNA-197 can significantly affect the biological behavior of OSCC cell,such as: proliferation,invasion and lymphatic metastasis.Mi RNA-197 might be a new potential target for OSCC diagnosis and therapy.
