| The increased incidence of myocardial infarction and the young for this trend and both more obvious.Percutaneous coronary intervention therapy(PCI)has good safety and operability and is the main means of treating myocardial infarction.However,due to the large dose of contrast agent during PCI surgery,contrast agent nephropathy(CIN)has become an important complication after PCI surgery,which has also attracted the attention of clinical workers.Grape seed polyphenols(GSP),a natural plant polyphenols extracted from grape seeds,is a kind of natural antioxidant with good water solubility and low toxicity.At present,it has been reported that GSP has anti-cardiovascular atherosclerosis,diabetes and hyperlipidemia effects,but the protective effect of contrast agent nephropathy has not yet been reported.Therefore,this paper mainly studies the effect of GSP on renal function protection in rats with acute myocardial infarction contrast agent nephropathy,and provides good protective means for clinical application.Objective: To study the effects of GSP orally administration on acute myocardial infarction contrast agent nephropathy in order to provide an effective new treatment approach for clinical medical workers.Methods: The 28 wistar male rats weighing around 200 g were randomly divided into 4 groups,the normal control group(Control,C),the acute myocardial infarction and the injection contrast agent group(AMI + CM),and the CM + AMI + GSP low dose group(low,L)and CM + AMI + GSP high-dose group(high,H),7 rat for each group.The diet,drinking water,mental state and fur color of the rats were observed.The rat AMI model was induced by preoperative ligation in the ventricle wall for paleness,and then we injected rats with iopromide(2 g/kg,i.v.)after ligation.The diet,drinking water and mental state,coat color and weight in rats were observed before operation 7 d to after operation 1 d.The indexes of serum creatinine(Scr)from extraction of vena cava blood and ROS in renal tissues from rat model were detected after 24 h execution.Haematoxilin-Eosin(HE)staining was performed to observe the pathological changes of kidney tissue and morphology;Immunohistochemical assay was to detectede caspase-3 expression in kidney tissues of rats.Western blotting assay was used to confirm caspase-3 expression in kidney tissues.Results: There was no significant difference in weight between the groups of rats with good preoperative status before operation and depressed after operation.In the AMI + CM group,serum creatinine(Scr)and reactive oxygen species(ROS)were obviously elevated,and after GSP gastric irrigation,both L and H groups showed a tendency to inhibit oxidative stress and showed a dose-dependence.Comparison with AMI + CM group showed a significant difference(P < 0.05);In AMI + CM group,the increased percentages of renal histopathology score and renal tubules apoptosis was also significant.However,after GSP was administration,the pathological changes of the kidneys in the L and H groups and the glomerular damage were relieved.The percentage of renal pathology score and renal tubule apoptosis decreased significantly in group H(P < 0.05).The results of Western blot also confirmed that compared with C group,the positive Caspaste-3 expression was higher in AMI + CM group,it was decreased in L group,and Caspaste-3 positive expression was significantly decreased in H group(P<0.05).Conclusions: GSP gastric lavage has a certain therapeutic effect on contrast agent nephropathy in rat models of acute myocardial infarction.Its pharmacological mechanism is mainly related to its good antioxidant and anti-apoptosis functions. |
