Objective:To detect the Sortilin-associated receptor-1(SORL1)gene and nucleotidebinding oligomerization domain-like receptor protein-3 in the peripheral blood of the amnestic mild cognitive dysfunction(aMCI)group and the control group(NC group).(NLRP3)Gene mRNA relative expression,explore the role of SORL1,NLRP3 gene in the pathophysiological process of aMCI patients,and provide a theoretical basis for further screening of biomarkers and therapeutic targets in the early diagnosis of aMCI patients.Methods:1.According to the criteria for admission,the patients aged 60-79 years old from the first hospital affiliated to Jiamusi University from December 2017 to December 2018 were enrolled.There were 32 patients in the aMCI group,13 males and 19 females.The age was 66.60±8.40 years old,and the NC group was 28 cases,16 males and 12 females,aged 62.06±1.89 years old.2.The inclusion of the aMCI group is the 2018 Chinese Diabetes and Cognitive Diagnostic Guidelines.The neuropsychological assessment methods used include the Simple Intelligence Mental Status Examination Scale(MMSE),the Montreal Cognitive Assessment Scale(MoCA),and the Auditory Word Learning Test(AVLT),Wired Test(TMT),Boston Named Test(BNT).3.All subjects were asked for general clinical data,brain MRI or CT examination,Hamilton Anxiety Scale(HAMA),Hamilton Depression Rating Scale(HAMD),Hachinski Ischemic Scale,Clinical Dementia Rating Scale(CDR),Activity of Daily Living Rating Scale(ADL).To rule out other diseases.All subjects received 5ml of fasting venous blood in the early morning of the next day.4.Real-time PCR(RT-PCR)was used to quantitatively analyze the SORL1 gene and NLRP3 gene,and the relative expression of SORL1 gene and NLRP3 gene was obtained.Statistical analysis of data was performed using SPSS 20.0 software.Results: The relative expression of SORL1 mRNA in aMCI group was significantly lower than that in NC group(P<0.05).The relative expression of NLRP3 gene mRNA in aMCI group was significantly higher than that in NC group(P<0.05).The SORL1 gene and NLRP3 gene in aMCI group There was a negative correlation between the relative expression levels(r=-0.984,P<0.05).Conclusions: SORL1 and NLRP3 genes are involved in the pathophysiological process of aMCI and have certain correlations.
