Objective:Primary liver cancer is a common malignant tumor in China.Its high incidence and low survival rate seriously endanger the safety of life and property of our people.Therefore,by exploring the mechanism of the occurrence and development of primary hepatocellular carcinoma,the selection of key sites may provide a choice for future treatment.The purpose of this study is to investigate the effect of TRPV4 channel protein on the oncological characteristics of hepatocellular carcinoma cells by analyzing the expression of TRPV4 channel protein in common benign and malignant liver diseases,and to reveal its possible role in the development of hepatocellular carcinoma.Methods:1.Collect the pathological specimens and clinical data of liver surgery for various diseases in the First Hospital of Shanxi Medical University from August 2012 to August2017.The expression of TRPV4 in common benign and malignant liver diseases was studied by immunohistochemical method,and the relationship between the expression of TRPV4 and the data of corresponding patients was analyzed.2.Further detection of TRPV4 gene and protein expression in Huh-7 and HepG-2hepatocellular carcinoma cell lines and comparison with normal human hepatocellular LO2 cell lines;regulation and consumption of protein expression in hepatocellular carcinoma cell lines by small interfering RNA(si-TRPV4)transfection and non-selective blocker ruthenium red blockade,and further detection of proliferation,invasion and metastasis of low protein expression hepatocellular carcinoma cell lines.Changes in migration and other aspects were compared with untreated hepatocellular carcinoma cell lines.Results:1.Immunohistochemistry showed that the expression of TRPV4 was different between hepatic hemangioma and cirrhosis(c~2=10.407,P=0.001).The expression of TRPV4 in hepatic hemangioma and hepatocellular carcinoma(c~2=14.005,P<0.001)was significantly different,suggesting that TRPV4 might play different roles in the pathogenesis of benign and malignant liver diseases.The expression of parahepatic tissue was significantly different(c~2=5.082,P=0.024),suggesting that it may play a role in the development of hepatocellular carcinoma.Immunohistochemical results combined with clinical data analysis showed that the difference of TRPV4 expression may be related to the degree of differentiation of tumors(P=0.025),and the difference of TRPV4expression may be related to the size of hepatic hemangioma(P=0.019).Due to the lack of data,further verification is needed.Small interfering RNA(si-TRPV4)transfection and ruthenium red(RuR)blockade were performed on Huh7 and HepG-2 cell lines respectively.Western blot results showed that both techniques inhibited the expression of TRPV4 channel proteins.CKK-8 and Transwell tests indicated that the difference of TRPV4 protein channel expression might play a role in the proliferation and migration of hepatocellular carcinoma cells.The displacement effect is particularly obvious.2.Small interfering RNA(si-TRPV4)transfection and ruthenium red(RuR)blockade were performed on Huh7 and HepG-2 cell lines respectively.Western blot results showed that both techniques inhibited the expression of TRPV4 channel proteins.CKK-8 and Transwell tests indicated that the difference of TRPV4 channel expression might play a role in the proliferation and migration of hepatocellular carcinoma cells,especially in the migration of hepatocellular carcinoma cells.Conclusion:TRPV4 channel protein is expressed in different benign and malignant diseases of the liver,and there is a significant difference in the expression of TRPV4 channel protein between malignant liver tumors and adjacent tissues,suggesting that TRPV4 channel protein may play a role in the development of hepatocellular carcinoma.Experiments on Huh7 and HepG-2 hepatocellular carcinoma cell lines suggest that the differential expression of TRPV4 protein channel may play a role in the proliferation and migration of hepatocellular carcinoma.Differential expression of TRPV4 channel proteins may promote the occurrence and development of hepatocellular carcinoma.The role of TRPV4 expression in the occurrence and development of hepatocellular carcinoma may contribute to the treatment and prognosis of primary hepatocellular carcinoma.However,further proteomics,genomics,animal and clinical trials are needed to confirm the possible role of TRPV4 channel proteins in hepatocellular carcinoma.
