Objective:The purpose of this study was to detect the expression of Ki67 protein and HAND2protein in different endometrial tissues,and to analysis the relationship between them and the pathological clinical features of endometrial carcinoma(EC).Then to investigate the significance in the occurrence and the development of endometrial carcinoma.Methods:Collecting pathological tissue 125 cases which had been removal of patients endometrial tissue by surgical operation and been manufactured paraffin specimens.They are classified into three groups and consist of normal endometrial tissue 30 cases(marked as group A),atypical hyperplasia endometrial tissue 20 cases(marked as group B),and endometrial carcinoma tissue 75 cases(marked as group C).We will detect the expression of Ki67proteinandHAND2proteinindifferentendometrialtissuesbyusing immunohistochemical SP method and analysis of the relationship between the two characteristic parameters with clinical cases respectively and the correlation between them.Using?~2 test to compare the differences among counting data in three groups and using Spearman method to analysis the correlation between Ki67 protein and HAND2 protein in endometrial carcinoma.Results:1.The Ki67 protein is mainly expressed in nucleus.The positive expression rate of Ki67 in group A is 23.33%,the positive expression rate of Ki67 in group B is 60%,the positive expression rate of Ki67 in group C is 66.67%,the expression of them showed a uptrend.Among the three groups,the difference was statistically significant(?~2=17.584,P=0.007<0.05).2.The HAND2 protein is mainly expressed in nucleus.The positive expression rate of HAND2 in group A is 90%,the positive expression rate of HAND2 in group B is 40%,the positive expression rate of HAND2 in group C is 25.33%.Among the three groups,the difference was statistically significant(?~2=41.021,P=0.000<0.05).3.In75 cases endometrial carcinoma,the positive expression of Ki67 protein was significantly different in tumor grade and staging respectively(FIGO stage),and the difference was statistically significant(P<0.05).But the expression discrepancy of Ki67 protein had no statistical significance(P>0.05)among the clinical parameters of patients with EC patients(lymph node metastasis\pathological type\depth of myometrium invasion).4.the expression of HAND2 protein was significantly different in staging respectively(FIGO stage)and pathological type,and the difference was statistically significant(P<0.05).But the expression discrepancy of HAND2 protein had no statistical significance(P>0.05)among the clinical parameters of patients with EC patients(tumor grade\lymph node metastasis\pathological type\depth of myometrium invasion).5.In endometrial carcinoma,the expression of Ki67 protein was associated with the expression of HAND2 protein,C_p=0.417.Conclusion:1.Among in group A\B and C,the expression of Ki67 protein is gradually increased.On the contrary,the expression of HAND2 protein shows a decrease.They may be involved in the occurrence and development of endometrial carcinoma.2.In 75 cases endometrial carcinoma,Ki67 protein is obviously high expression,and HAND2 protein is significantly low expression.The expression of the two is correlated,and it is speculated that they have a certain clinical value to the early diagnosis EC.
