| Objective:To analyze the association between high mobility group box1(HMGB1)gene polymorphisms and hepatitis B virus(HBV)-related acute liver failure(ALF),and to explore its possibility of early prediction of ALF susceptibility.Methods:30 patients with HBV-related ALF were collected as the case group from several hospitals in Shanxi Province from September 2013 to August 2018,during the same period60 patients with acute hepatitis B and 60 normal persons were collected as the control group from the first Hospital of Shanxi Medical University.The genotypes of rs1045411,rs3742305,rs2249825,rs1412125,rs9508752 and rs2892547 in HMGB1 gene were detected by the first generation sequencing method.χ 2 test and non-conditional Logistic regression were applied to analysis results.Results:In case group and control group,the genotype and allele frequency of 6 SNPs were in line with the Hardy-weinberg equilibrium(HWE)(P > 0.05).The C allele frequency of rs1412125 in HBV-related ALT group was higher than that in acute hepatitis B group(χ2 =4.043,P=0.044),there was no significant difference in other SNPs’ genotype distribution and allele frequency between case group and control group(P > 0.05).Excluding age and sex factors,in codominant and dominant modes,the susceptibility to HBV-related ALF was increased in normal individuals with rs1412125 CC or CC+CT genotype(OR=4.829,95%CI : 1.018-22.903,Codominant model;OR=3.289,95% CI : 1.108-9.761,Dominant model);acute hepatitis B patients with rs9508752 AG or AG+GG genotype were more likely to develop to HBV-related ALF(OR=3.934,95% CI:1.061-14.583;OR= 4.129,95%CI:1.219-13.985).Conclusion:The polymorphisms of rs1412125 and rs9508752 of HMGB1 gene were associated with the risk of HBV-related ALF.The polymorphisms of rs1045411,rs3742305,rs2249825,rs2892547 may not be associated with HBV-related ALF. |
