[Purpose]High-mobility group protein1(HMGB1)is a non-histone nuclear protein whose function depends on cellular location. HMGB1modulates a variety of important cellular processes.within cell,It is involved in nucleosome stabilization and transcription regulation. Whereas,released HMGB1acts as a cytokine that can promote inflammation response and contribute to the pathologic progression of infectious and inflammatory disorders.Extracellular HMGB-1were related with a variety of acute and chronic inflammation as the inflammatory factor.Accumulated evidences show that serum HMGB1expression is increased in patients with viral hepatitis,Howerver,the relationships between serum levels of HMGB1and different kinds of viral hepatitis,liver sclerosis and liver cancer in patients have not been elucidated.This paper aims to determine serum HMGB1levels in various liver diseases using the ELISA,study correlationship between the severity and serum HMGB1levels,and analyse the relationship between HMGB1level and other lab diagnostic parameters in patient with viral hepatitis.[Methods]1. Qualitative ELISA-kit was used to detect serum HMGB1level of patients and control group.2.Automatic biochemical analysis(TOSHIBA-120FR) was used to detect relative biochemical parameters of both patients and control groups.3.Using SN-695A RIA y measuring instrument to measure parameters of hepatic fibrosis of both patients and control groups4.Using automatic coagulation analysis machine sysmex CA-530to measure Prothrombin time of both patients and control groups.5.Using Electrochemical automatic immune analysis machine to detect hepatocellular carcinoma-related indicators of both patients and control groups[Results]1.The serum HMGB1levels of patients with chronic hepatitis, severe hepatitis, hepatocirrhosis,liver cancer are higher than that of those of normal control group, HMGB1expression was significantly increased in severe hepatitis patients in comparison with that in chronic hepatitis B patients,hepatocirrhosis and liver cancer patients(P<0.01); serum HMGB1levels of patients with hepatocirrhosis,liver cancer are higher than that of Chronic hepatitis group(P<0.01),but there is no difference between hepatocirrhosis group and liver cancer group(P>0.05).2.The serum HMGB1level increase following the aggravation of liver damage in patients with CB. The serum HMGB1level in patients with chronic severe hepatitis is significantly higher than that in the Chronic moderate hepatitis and chronic mild hepatitis(P<0.01).ALT,TBIL,ALB,ASTm,HA in patients with chronic severe hepatitis are significantly higher than in the chronic moderate hepatitis and chronic mild hepatitis(P<0.01), The difference also was observed between the chronic moderate hepatitis and chronic mild hepatitis (P<0.01).3.The level of HMGB1in patient with hepatocirrhosis significantly increases with the aggravation of liver fibrosis. The serum HMGB1in child c group is higher than in child A and child B groups (P<0.01).ALT,AST,TBIL,AST-m,GLDH,HA,LN,Pâ…¢P in child c group are higher than in child A and child B groups(P<0.01), LN level has no different between child A and child B group (P>0.05). IV level has no different in three groups(P>0.05).4.There is positive correlation among serum HMGB1expression and ALT,TBIL,PT,AST-m,GLDH,GPDA,HA,LN,Pâ…¢P expression in patients with chronic hepatitis(HMGB1with, TBIL,AST-m,GPDA,HA,LN and PIIIP P<0.01; HMGB1with ALT, PT and GLDH P<0.05). Serum HMGB1level in the patient with severe hepatitis has a positive correlation with TBIL,AST-m,PT,LN,Pâ…¢P(HMGB1with TBIL,LN,PIIIP P<0.01:HMGB1with AST-m,PT P<0.05).Serum HMGB1level in hepatocirrhosis patients has a positive correlation with TBIL,AST-m,PT,LN, PIIIP (HMGBl with TBIL,ASTm,HA P<0.01; HMGB1with LN P<0.05). But there is negative correlation between HMGB1expression and ALB expression (P<0.01) in hepatocirrhosis patients. Serum HMGB1level has a positive correlation with TBIL, GLDH,HA,IV,PIIIP,CEA,CA199in liver cancer patients (TBIL,â…£,Pâ…¢P,CEA, CA199P<0.01; GLDH,HA P<0.05)[conclusion]The results show serum HMGB1level of patients with liver disease are higher than those of control groups, and serum HMGB1level has a positive correlation with ALT,TBIL,ASTm,HA,LN,Pâ…¢P,and serum HMGB1level is closely associated with the aggravation of liver damage and liver fibrosis. These results suggest that HMGB1may be involved in the pathogenesis of hepatitis B, liver fibrosis and hepatoma, and could be biomarker for those diseases.
