Objective The incidence of lung cancer in the world is very serious,and non-small cell lung cancer is the most common type of lung cancer.It is also a hot spot of current research.With the renewal and development of technology and medical technology,the cure rate of non-small cell lung cancer has changed,but its pathogenesis is still unknown,so it is necessary to continue to explore and study the mechanism of occurrence,development and metastasis of non-small cell lung cancer.Seek more specific molecular diagnostic markers and gene therapy targets.LINC00261 has biological functions in a variety of cancers,but the mechanism of action in cancer is not clear.Therefore,this study uses bioinformatics methods to mine database-related data to predict and analyze the mechanism of action of LINC00261 in non-small cell lung cancer.It is expected to provide information for the signaling pathway research of lung cancer,find new directions and means for the clinical treatment of lung cancer,and provide theoretical basis and pathological mechanism for the basic research of lung cancer.Methods By analyzing the gene chip data of GEO and TCGA databases,analyzing and screening differentially expressed genes,enriching the pathways by GO function and KEGG,and verifying the correlation of LINC00261 target genes in TCGA database.Results The results of screening differentially expressed genes showed that there were 864 differentially expressed genes,of which 425 were up-regulated genes,accounting for 49.19%,and 439 were down-regulated genes,accounting for 50.81%.Down-regulated IncRNA has the largest down-regulation of LINC00261 and is much larger than other lncRNAs.The GO functional classification analysis showed that most of the genes belong to the cellular components and the originals in the biological process,while the genes that perform the molecular functions are relatively few.And differentially expressed genes are mainly involved in cell proliferation and mitosis.KEGG pathway enrichment analysis,cell cycle in differentially expressed genes,P53 signaling pathway,etc.are active in non-small cell lung cancer tissues than normal paracancerous tissues.The correlation verification of LINC00261 target gene in the TVGA database showed that the target gene of LINC00261 was also Forkhead box protein A2(FOXA2)in non-small cell lung cancer patients,the LINC00261 gene was significantly down-regulated,and FOXA2 was significantly up-regulated.Conclusion In this paper,bioinformatics methods were used to analyze non-small cell lung cancer cases in the database.The results showed that LINC00261 is significantly low expression in non-small cell lung cancer and has the function of tumor suppressor gene,which can inhibit the growth,metastasis and growth of non-small cell lung cancer cells.Invasion.LINC00261 reduces the growth,metastasis and invasion of tumor cells by activating FOXA2 expression,thereby inducing cells to express TGF-?1 and inhibiting cell EMT.
