Objective Purpose to establish a real-time fluorescent quantitative PCR(policy real-time polymerase chain reaction,RT-qPCR)method was used to detect the expression levels of miR-1910-3p in serum of breast cancer patients and healthy control group and serum exosome sources,and to analyze the correlation between miR-1910-3p and clinical pathological features,so as to explore the feasibility of it as a serum marker for the early diagnosis of breast cancer.The clinical diagnostic value of miR-1910-3p compared with CEA and CA153,the traditional tumor markers,was also discussed.Method Random charge the second hospital affiliated to suzhou university without any cancer treatment serum specimens in serum specimens of breast cancer patients and healthy controls all 25 cases,exosome extract serum source,RT-qPCR method is used to test each serum specimens of miR-1910-3p and serum exosome source miR-1910-3p relative expression,at the same time using automatic chemiluminescence detection of CEA and CA153 in serum specimens of each content.In this study,receiver operating characteristic curve(ROC)was used to evaluate the diagnostic performance of miR-1910-3p,CEA,CA153 and combined diagnosis.Res?Lts The relative expression of miR-1910-3p were 1.740(0.417,3.797)and 0.689(0.401,2.312)in the serum of the breast cancer patients and the healthy control group by RT-qPCR,and the differences were not statistically significant(P=0.1936).CEA expression levels were 1.420(0.960,2.390)ng/mL and 1.110(0.730,2.155)ng/mL,respectively,with no statistical significance(P=0.1904).The expression levels of CA153 were respectively 10.370(7.485,15.530)IU/mL and 7.650(6.665,10.530)IU/mL,with statistically significant differences(P=0.048).The relative expression of miR-1910-3p from exosome in the experimental group and control group were 6.763(2.978,13.118)and 1.386(0.738,2.367),respectively,suggesting that the relative expression levels of miR-1910-3p from exosome in the breast cancer patients were significantly higher than those in the healthy control group(P<0.001).The content of miR-1910-3p from exosome was significantly positively correlated with breast cancer size,with statistically significant differences(P=0.035),and had no significant correlation with age,tumor pathological type,lymph node metastasis or tumor stage(P>0.05).The diagnostic efficacy of miR-1910-3p and CA153 from exosome was analyzed by ROC curve,and Youden's index was taken as the boundary value.The area under the ROC curve(AUC)of the former was 0.8864(95%CI 0.7937-0.9791),the critical value was 2.144,and the sensitivity and specificity were 88%and 76%,respectively.The area under the ROC curve was 0.6624(95%CI 0.5104?0.8144),the critical value was 8.395U/mL,and the sensitivity and specificity were 68%and 60%,respectively.A combination of serum exosome-derived mir-1910-3p and CA153 had a 96 percent sensitivity to diagnosis of breast cancer.Conclusions Serum exosom-derived mir-1910-3p plays a role as a non-invasive biomarker in the auxiliary diagnosis of breast cancer,and the diagnostic efficacy of exosom-derived mir-1910-3p for breast cancer is better than that of traditional tumor marker CA153.This study provides a new idea for the diagnosis of breast cancer.
