objective:The expression of HSP70 in placental tissues and serum of normal pregnancy and preeclampsia pregnant women were analyzed.Through the cell experiments,we detection the expression of HSP70,Caspase-3,and the NF-?B.Further more,we discusses on the pathogenesis of preeclampsia and possible treatment options on clinical.Methods:1.Clinical research:A total of 70 samples were collected in Wuxi Nanzha hospital of Jiangsu province and Hospital of Suzhou from February 2017 to May 201 8.There were 15 cases of preeclampsia in the placental sample and the normal control group,36 cases of preeclampsia in the serum sample and 34 cases in the normal control group.The content of HSP70 was determined by immunohistochemistry and ELISA2.Cell experiments:Human Umbilical Vein Endothelial Cells(HUVEC)hypoxic ischemia model was established to detect the expressions of HSP70,Caspase-3 and NF-?B And we up-regulated and down-regulated in preeclampsia cell model,to further verified and the analyze the correlation between HSP70,Caspase-3 and NF-?B.Results:1.Compared with normal pregnancy,the expressions of HSP70 in preeclampsia patients were significantly increased both in the placenta and serum(P<0.01).2.In the ischemic and hypoxic cell model,the expression of HSP70,Caspase-3 and NF-?B was higher than that of normal cultured cells(P<0.01).3.After the addition of HSP70 agonists,the expression of Caspase-3 and NF-?B both increased.,and after the addition of HSP70 inhibitors,the expression of Caspase-3 and NF-?B both decreasedConclusion:1.The expressions of HSP70 of preeclampsia patients should be significantly increased in placenta and serum,suggesting that expression of HSP70 increased after oxidative stress induced by oxidative stress.2.HSP70 promotes apoptosis by increasing the activity of Caspase-3;at the same time,increasing the expression of NF-?B can promote the pathogenesis of preeclampsia...
