1.Detection of E-cadherin in rats with acute pancreatitisObjective:To explore the diagnostic value and mechanism of E-cadherin in the acute pancreatitis.Methods:Twenty-four male SD rats were randomly divided into the control group,the MAP group and the SAP group.The animal model was established by intraperitoneal injection of L-arginine.After 24 hours,the serum levels of amylase,WBC,CRP,PCT and E-cadherin were tested.The pancreas tissue was taken for histopathological score under microscope.The expression of E-cadherin at protein level in pancreatic tissue was detected through Western Blot.Results:The histological score of pancreas and serum amylase were significantly higher in the MAP and SAP groups than those in control group(P<0.05),and higher in SAP group than in MAP group(P<0.05).The detection value of WBC were significantly higher in the MAP and SAP groups than those in control group(P<0.05),but there was no significant difference between SAP group and MAP group(P>0.05).The detection values of CRP and PCT were significantly higher in the MAP and SAP groups than those in control group(P<0.05),and higher in SAP group than in MAP group(P<0.05).Using ELISA,the serum E-cadherin in MAP group and SAP group were significantly higher than that in control group,and there was significant difference between MAP group and SAP group(P<0.05).The result of Western Blot was similar to the result of ELISA.Conclusion:The experiment demonstrated that E-cadherin had diagnostic value in the acute pancreatitis.2.Detection of E-cadherin in patients with acute pancreatitisObjective:To explore the early diagnostic value of E-cadherin in the patients with acute pancreatitis.Methods:Clinical data of 76 patients with AP were collected;the serum levels of E-cadherin,CRP,PCT,WBC and amylase were measured at different sampling points(12 hours,24 hours and 48 hours).The patients were divided into the MAP group,MSAP group and SAP group;then compared the diagnostic value at different time points.Results:The detection value of serum amylase was no significant difference between MAP group,MS AP group and SAP group at 12 hours after the onset of the disease(P>0.05);there were significantly higher in the MSAP and SAP groups than those in MAP group after 24 hours(P<0.05),and higher in SAP group than in MSAP group(P<0.05);there were significantly higher in the MSAP and SAP groups than those in MAP group after 48 hours(P<0.05),but there was no significant difference between SAP group and MSAP group(P>0.05).The detection value of WBC were significantly higher in the MSAP and SAP groups than those in MAP group after 12hours and 24 hours(P<0.05),but there was no significant difference between SAP group and MSAP group(P>0.05);however there was significantly higher in the SAP group than those in MAP group and MSAP group after 48 hours(P<0.05),but there was no significant difference between MSAP group and MAP group(P>0.05).The detection values of CRP and PCT alkie were no significant difference between MAP group,MSAP group and SAP group at 12 hours after the onset of the disease(P>0.05);but there were significantly higher between MAP group,MSAP group and SAP group after 24 hours and 48 hours.The serum E-cadherin in MSAP group and SAP group were significantly higher than that in MAP group,and there was significant difference between MSAP group and SAP group at different sampling points(12 hours,24 hours and 48 hours)(P<0.05).Conclusion:The serum amylase and WBC were not directly proportional to the severity of the disease and could not predict the severity of the disease.CRP and PCT have clinical value in judging the severity of the disease at 24 hours after the onset of the disease,but their predictive value is limited in the early stage of the disease(before 24 hours).But the detection of E-cadherin has important clinical value in judging the severity of AP in early stage.Summary:Through detection the serum E-cadherin by ELISA and detection of protein expression in pancreatic tissue by Western blotting in AP rat model;and detection of E-cadherin in AP patients with different severity at different time points.They are suggested that E-cadherin is highly expressed in the early stage of AP,which has important clinical value in judging the severity of AP.
