Objective:This study is mainly to analyze the molecular characteristics of angiotensin-induced aortic aneurysm formed by ascending aortic aneurysm induced by angiotensin-aortic aneurysm by and aortic arch constriction and to explore their respective molecular mechanisms.Methods:The anorectal or abdominal aorta was induced by angiotensin combined with a high-fat diet.The aortic diameter of the mouse was determined by cardiac ultrasonography.Transcriptional analysis of the aorta-forming site and normal aortic tissue of the same mouse to obtain differentially expressed genes,and select genes with differential expression more than double,and then use the GO enrichment analysis and literature search methods from the above genes.Screening for genes involved in aortic aneurysm formation.The ascending aortic aneurysm was formed by aortic arch constriction surgery,and the aortic diameter of the mouse was measured by cardiac ultrasonography to determine the successful model of the ascending aorta in the mouse.The ascending aorta and the normal aortic tissue of the same mouse were transcribed.Spectral analysis to obtain differentially expressed genes.Select genes with more than double differential expression,and then select genes involved in aortic aneurysm formation from the above genes by GO enrichment analysis and literature search.Results:In the experimental group of angiotensin combined with high-fat diet,aortic aneurysms were formed in the ascending aorta or abdominal aorta.The average aortic diameter was 2.0±0.035mm,and the aortic diameter of the control mice was 1.25±0.024 mm.The differentially expressed genes of more than 2 times in the ascending aorta and abdominal aorta were screened.The genes involved in aortic aneurysm formation were obtained by GO enrichment analysis and literature search:Kdm5d(histone demethylase)expression 39.3 times higher,Ccl8(chemokine CCL8)expression increased 4.9 times,Ptgs2(prostaglandin epoxide synthetase 2)increased 3.5-fold,and Rgsl(recombinant human G-protein signal regulator 1)increased 3.4.Times.These genes are involved in the involvement of atherosclerosis,chemotaxis of inflammatory factors,and regulation of endothelial cell function,leading to the formation of aortic aneurysms.The experimental group formed by aortic arch constriction surgery formed an aortic aneurysm in the ascending aorta with an average aortic diameter of 2.0±0.026 mm,and the control group had aortic diameter of 1.00±0.035 mm.The differentially expressed genes in the ascending aorta were more than 2-fold and the genes involved in aortic aneurysm formation were analyzed by Pathway.The expression of Illm(interleukin-1 receptor antagonist)was increased by 48.5 times,Mmp12(human metal)The expression of matrix protease 12)was increased by 10.5 fold,the expression of C3arl(complement 3a receptor 1)was increased by 10.1 fold,the expression of Tnf(tumor factor)was increased by 9.2 fold,and the expression of TIMP-1(tissue matrix metalloproteinase inhibitor-1)was expressed.8.2 times higher.These gene functions include chemotaxis of inflammatory factors,degradation of extracellular matrix,infiltration of macrophages,etc.,leading to the formation of aortic aneurysms.Conclusions:Through gene chip detection,GO enrichment analysis,angiotensin combined with high-fat diet model and ascending aortic aneurysm model,the above-mentioned high-expression genes are involved in signal transduction,chemokine chemotaxis,and cell matrix degradation.These highly expressed genes are involved in the formation of aortic aneurysms.
