Objective:To study serum miRNA expression profiles of Patients with Gallstones and healthy people,and explore their different expression of miRNA.Methods:We prospectively collected the serum of 25 patients with gallstones and 25 healthy people in our hospital.The miRNA expression profile was established by miRNA sequencing technology.The differential miRNA was verified by qPCR and its biological functions were predicted and analyzed by the bioinformatics technology.The quantitative data were described by mean ± SD,and were compared by T-test.Results:There was no significant difference in general information between two groups.The sequences of serum miRNA expression profiles in the study group and the control group were 14899245 and 11783121,respectively,and the miRNA were 686 and 633,respectively.A total of 16 miRNAs were significantly different expression between two groups,which were down-regulated.qPCR confirmed confirmed the differences of miR-1228,miR-1249,miR-3614,and miR-766 expression,which accorded with the result of miRNA sequencing.There were significant difference between two groups in miR-1228,miR-3614 and miR-766(t=3.57,3.49,3.76,P<0.05),but not in miR-1249(t=3.70,P>0.05).GO analysis showed that the target genes were enriched in biological process-cellular process,biological regulation and metabolic process,cellular component-cell part,organelle and organelle part and molecular function-binding and catalytic activity.KEGG analysis showed that target genes were enriched in cAMP signaling pathway,oxytocin signaling pathway and circadian entrainment.Conclusion:The expression profile of serum miRNA is different between patients with gallstones and healthy people.The differential expression serum miRNAs may regulate cholesterol metabolism and play an important role in the formation of gallstones.GO and KEGG analysis show that target genes of differential expression serum miRNAs are involved in lipoprotein particle binding,protein kinase activity,bile secretion pathway and thyroid hormone signaling pathway,which are associated with the formation of gallstones.
