Objective:To observe the Yijing Tongluo Formula can improve the defocus-induced myopia of guinea pig retinal free dopamine content,prevent lengthening of Axis Length and thinning of Choroid Thickness,and inhibite myopia progression.To provide theoretical basis for the clinical application of this prescription.Methods:The research devided into two parts.The first prat is Yijing Tongluo Formula infects the defocus-induced myopia of guinea pig in eye structure.Selection two-week-old guinea pigs,40,randomly divided into 4 groups,is Chinese medicine group,Atropine control group,Model control group and Normal control group.The Chinese medicine group was given Yijing Tongluo Formula gavage 2 ml/time/day and defocus-induced(-5D);The Atropine control group was given 0.01%Atropine eye drops 1 drop/time/night and defocus-induced(-5D);The Model control group was only given defocus-induced(-5D);The Normal control group was not given any treatment;Right Eye for defocus induced.Intervention 4 weeks.All guinea pigs were examined by mydriasis,optometry and A super measurement before and after intervention.One-way ANOVA analysis was performed for Diopter,Axis Length,Anterior Chamber Depth,Len Thickness and Vitreous Length before and after intervention among different groups,and paired sample t test was performed for Diopter,Axis Length,Anterior Chamber Depth,Len Thickness and Vitreous Length before and after intervention in every groups.Choroid Thickness was measured by HE staining and NIS-Elements image analysis system,and one-way ANOVA was performed after intervention among different groups.The correlation among the after intervention Diopter and Axis Length,Vitreous Length,Anterior Chamber Depth,Len Thickness,Choroid Thickness were analyzed.The second part:To explore the effect of Yijing Tongluo Formula on retina DAT content in myopia guinea pigs.After 4 weeks of intervention,eyeball removal,fixation,frozen sections were made.The expression of DAT in retina was detected by immune histochemical method and the expression of DAT positive cells in retina was calculated by NIS-Elements image analysis system,and one-way ANOVA was performed inter-group comparison.Results:Inter-group comparison:There were no significant difference in Diopter,Axial Length,Vitreous Length,Anterior Chamber Depth and Len Thickness between the four groups before intervention(P>0.05);There were significant differences in Diopter,Axial Length and Vitreous Length between the four groups after intervention(P<0.05),and there were no significant difference in Anterior Chamber Depth and Len Thickness(P>0.05).There was no significant difference in Choroid Thickness between the four groups after intervention(P>0.05),except the difference between the Chinese medicine group and the Atropine control group(P<0.05),and there were significant difference between the other two groups(P<0.05).There was no significant difference in the number of DAT positive cells between the four groups after intervention(P>0.05),except the difference between the Chinese medicine group and Atropine control group(P>0.05),and there were significant difference between the other two groups(P<0.05).Group comparison:There were significant differences in Diopter,Axial Length and Vitreous Length before and after intervention in each group(P<0.05).There were no significant difference in Anterior Chamber Depth and Len Thickness(P>0.05).Correlation analysis:The refraction of defocused moderate myopia guinea pigs was negatively correlated with the Axial Length and Vitreous Length,but not with Anterior Chamber Depth,Len Thickness and Choroid Thickness.Conclusion:1.With the increase of Diopter,the defocus-induced myopia of guinea pig Axial Length and Vitreous Length increased significantly,the Choroid Thickness became thinner,and the changes of Anterior Chamber Depth and Len Thickness were not obvious.The moderate myopia Diopter is closely related to the Axial Length and Vitreous Length,but not to the depth of Anterior Chamber Depth,Len Thickness and Choroid Thickness.2.With the deepening of myopia,the expression of DAT in retina decreased,that is,the expression of free DA decreased.3.Yijing Tongluo Formula can inhibit the progress of defocus myopia in guinea pigs by inhibiting retinal DA content.4.Yijing Tongluo Formula and 0.01%Atropine eye drops can inhibit the progress of defocus myopia in guinea pigs.In terms of Diopter and Axial Length,0.01%Atropine eye drops has better effect than Yijing Tongluo Formula.but there was no difference in Choroid Thickness and free Dopamine content between the two groups.
